摘要
目的 探究神经胶质瘤患者外周血清中microRNA-21的表达水平,并运用生物信息学方法 预测其靶基因及功能.方法 选取神经胶质瘤患者53例作为试验组,另选取同期健康人群50例作为对照组.采用RT-PCR技术检测两组外周血清中microRNA-21的相对表达量,采用UCSC数据库分析microRNA-21在人类基因中的位置及保守性,采用Targetscan数据库和CoMeTa数据库联合预测microRNA-21的靶基因并进一步分析其靶基因的功能富集和信号通路富集情况.结果 试验组外周血清中microRNA-21的相对表达量(2.53±0.95)高于对照组(1.05±0.64),差异有统计学意义(P<0.05);microRNA-21定位于人染色体17q23.1上,具有高度保守性;生物信息学分析显示microRNA-21的潜在靶基因包括BCL2、SOCS3、MEF2C、PP3CA等146个;功能富集(GO)分析显示microRNA-21的靶基因主要富集于RNA合成代谢、细胞增殖凋亡、转录、磷代谢、细胞信号传导等过程中,而KEGG Pathway分析显示microRNA-21靶基因主要富集于丝裂原活化蛋白激酶(MAPK)信号通路、神经营养素信号通路、Janus激酶-信号转导与转录激活子(JAK-STAT)信号通路、细胞因子-细胞因子受体相互作用、趋化因子信号通路、Toll样受体信号通路、T细胞受体信号通路、细胞凋亡信号通路、转化生长因子(TGF)-β信号通路等通路中.结论 神经胶质瘤患者外周血microRNA-21高表达,并通过调控多条信号通路中多的种靶基因进而参与导致神经胶质瘤的发病.
Objective To determine the expression level of microRNA-21 in peripheral blood of patients with neuroglioma and predict its target gene and its function.Methods 53 patients with neuroglioma admitted to hospital were selected into experimental group(NG group),and 50 healthy people in the same period were selected into control group(Control group).RT-PCR was used to detect the relative expression of microRNA-21 in peripheral blood of the two groups.The location and conservation of microRNA-21 in human genes were analyzed by UCSC database.The target scan database and CoMeTa database were used to predict microRNA-21.The target gene was further analyzed for functional enrichment of its target gene and signal pathway enrichment.Results The relative expression of microRNA-21 in peripheral blood of patients with neuroglioma(2.53±0.95)was significantly higher than that of the control group(1.05±0.64),the difference was statistically significant(P<0.05);microRNA-21 located at human chromosome 17q23.1 was highly conserved;bioinformatics analysis showed that the potential target genes of microRNA-21 included 146 genes such as BCL2,SOCS3,MEF2C and PP3CA.GO analysis showed that the target gene of microRNA-21 was mainly enriched in RNA anabolism,cell proliferation and apoptosis,transcription,phosphorus metabolism,cell signaling,etc.,while KEGG Pathway analysis showed that the microRNA-21 target gene was mainly enriched in MAPK signal pathway,neurotrophin signaling pathway,JAK-STAT signaling pathway,cytokine-cytokine receptor interaction,chemokine signaling pathway,Toll-like receptor signaling pathway,T cell receptor signaling pathway,axon guidance signaling pathway,cell apoptotic signaling pathway,TGF-βsignaling pathway and other processes.Conclusions The expression of microRNA-21 in peripheral blood of patients with neuroglioma is high,and it could lead to the pathogenesis of neuroglioma by regulating multiple target genes and participating in multiple signaling pathways.
