摘要
目的探讨Tax1结合蛋白1(TAX1BP1)对B细胞增殖、活化、免疫球蛋白分泌等功能的影响。方法利用免疫磁珠法分选小鼠脾脏CD19^+B细胞,转染siRNA下调TAX1BP1的表达;并利用TAX1BP1慢病毒感染B细胞株Raji细胞系建立稳定下调TAX1BP细胞系。体外用LPS刺激B细胞后通过CCK8、流式细胞术和ELISA法分别评价TAX1BP1对B细胞增殖、B细胞表面共刺激分子表达、细胞周期以及免疫球蛋白分泌的影响。结果与对照组相比,敲低TAX1BP1促进了B细胞的增殖(P=0.008)及Raji细胞的增殖(P<0.0001),上调了B细胞表面活化分子CD69和CD80的表达(P<0.001),促进了免疫球蛋白lgG及lgM的生成(P<0.05)。结论TAX1BP1可以通过抑制B细胞增殖、活化、抗体产生等多方面调节B细胞功能。
This study was performed to investigate the role of Tax1-binding protein(TAX1BP1)on B cell fuction,including proliferation,activation,and immunoglobulin secretion.Freshly purified spleen CD19^+B cells from mouse were extracted and transfected with siRNA to down-regulate TAX1BP1 expression,then cultured in presence of LPS.TAX1BP1 lentivirus was used to infect the B cell line Raji cells to knockdown its expression.B cells were stimulated with LPS in vitro,and the cell proliferation,costimulatory molecules expression,and immunoglobulins secretion were assayed by CCK8,FCM and ELISA,respectively.Data showed that knocking down TAX1 BP1 in B cells promoted its proliferation,and this phenomenon also happened in Raji cells.Furthermore,down-regulating TAX1BP1 in B cells increased the expression of CD69 and CD80 molecules,and promoted immunoglobulin lgG and lgM secretion.In conclusion,TAX1BP1 can regulate B cell function by restricting its proliferation,activation and immunoglobulin secretion.
