摘要
通过体外细胞实验和动物模型实验,研究在食物过敏发生中发挥作用的Notch配体,并进一步明确其重要作用。结果表明:细胞模型中,鸡蛋过敏原卵清白蛋白显著增强了骨髓源树突状细胞上Notch配体Jagged1的表达,Jagged1通过上调Gata-3的表达促进Th2型细胞分化。小鼠过敏模型中,Jagged1-Fc融合蛋白显著增强了小鼠体内Th2型细胞因子分泌,增强了Th2型免疫反应,进而加重小鼠的过敏反应。因此,Notch配体Jagged1通过诱导Th2型免疫反应促进食物过敏的发生。
The role of Notch ligand Jagged1 in food allergy was explored in a cell model and an animal model.The results showed that ovalbumin enhanced the expression of Notch ligand Jagged1 on bone marrow-derived dendritic cells significantly.Jagged1-Notch signaling induced Th2 cell differentiation through enhancing the expression of Gata-3.In addition,treatment of Jagged1-Fc enhanced allergic responses significantly through promoting Th2 cell differentiation and Th2 cytokines production in mice.Jagged1-Notch signaling induced Th2 cell differentiation,and played a positive regulatory role in the sensitization phase of food allergy.
