摘要
目的通过体外研究明确GPD1L(甘油三磷酸脱氢酶-1类基因)基因对口腔鳞癌细胞系增殖、迁移及侵袭等生物学表型的影响。方法构建GPD1L过表达稳定转染细胞系,采用CCK8生长曲线、细胞划痕实验、重建基底膜侵袭实验和蛋白免疫印迹实验,明确GPD1L过表达前后口腔鳞癌细胞系增殖、迁移、侵袭能力及相关蛋白表达的变化趋势。结果口腔鳞癌细胞系WSU-HN6、Cal27和Tca83在GPD1L基因过表达后4~5天,出现明显增殖抑制及细胞周期S期阻滞。细胞划痕实验和重组基底膜侵袭实验发现,GPD1L基因过表达明显抑制口腔鳞癌细胞系Cal27和Tca83的迁移和侵袭能力。Western blotting实验证实GPD1L在口腔鳞癌细胞系中过表达可上调PHD2表达,并下调HIF-1α和Cyclin A2蛋白表达。结论GPD1L过表达可以抑制肿瘤细胞增殖、迁移、侵袭及细胞周期阻滞,从而影响口腔鳞癌细胞系体外生物学表型。
Objective To investigate the effect of GPD1L gene(glycerol-3-phosphate dehydrogenase 1-like)on the proliferation,migration and invasion of oral squamous cell carcinoma cell lines in vitro.Methods Based on the establishment of stable cell lines with GPD1L overexpression,CCK8 growth curve,wound healing assay,transwell test and western blot were used to determine the change trend of proliferation,migration,invasion ability and related protein expression of oral squamous cell carcinoma cell lines.Results Oral squamous cell carcinoma cell lines WSU-HN6,Cal27 and Tca83 showed obvious proliferation inhibition and s-phase cell cycle arrest which started 4 to 5 days following GPD1L gene overexpression.Wound healing assay and transwell test showed that overexpression of GPD1L gene significantly inhibited the migration and invasion ability of Cal27 and Tca83 cell lines.Western blotting assay confirmed that GPD1L overexpression in oral squamous cell carcinoma cell lines could up-regulate PHD2,and down-regulate HIF-1α and Cyclin A2 protein expression.Conclusion Overexpression of GPD1L can inhibit the proliferation,migration,invasion and cell cycle arrest of tumor cells,thus affecting the biological phenotype of oral squamous cell carcinoma cell lines in vitro.
