肉碱对液态保存猪精子质量和受精能力的影响 被引量:3

Effects of L-carnitine on sperm quality and fertilization ability during liquid storage of boar semen

摘要 探究肉碱对液态保存猪精子质量的影响,主要检测参数包括猪精子的活力、质膜完整性、抗氧化性(总抗氧化活力、活性氧含量和脂质过氧化水平)、ATP含量、线粒体活性和体外受精能力。结果显示,在保存液中添加肉碱可以有效维持猪精子的质量,如精子的活力和质膜完整性。同时发现,肉碱主要通过提高猪精子的抗氧化活力、提高ATP含量以及线粒体活性来维持猪精子质量。此外,本试验通过检测肉碱对精子受精能力和发育潜能的影响发现,添加肉碱可以增加精子结合到透明带的比例。结果表明,在精液保存液中适当添加肉碱可以改善在17℃液态保存的猪精子的质量。 Artificial insemination is becoming an increasingly prevalent in the boar industry.Therefore,it is necessary to prolong preservation of semen.The aim of this study was to evaluate the effect of the supplement L-carnitine to semen extender on sperm quality during boar sperm preservation.Parameters including motility,membrane integrity,antioxidant parameters(t-ROS,T-AOC and MDA)of boar sperm as well as ATP content,mitochondria activity were estimated and in vitro fertilizing ability of preserved boar sperm were also investigated.These data demonstrated that supplementation with L-carnitine could effectively maintain boar sperm quality such as sperm motility and membrane integrity.Besides,these data showed that L-carnitine had positive effects on boar sperm quality mainly through improving antioxidant capacities and enhancing ATP content and mitochondria activity.Interestingly by assessing the effect of L-carnitine on sperm fertility and developmental potential,these findings showed that the extender containing L-carnitine could improve sperm quality and increase the number of sperms bounding to zona pelucida.These findings suggested that the proper addition of L-carnitine to the semen extender could improve boar sperm quality during liquid storage at 17℃.
