摘要
目的:观察姜黄素改善大鼠肾脏缺血再灌注的作用机制。方法:45只雄性SD大鼠随机分为对照组、缺血再灌注模型组、姜黄素预处理组,比较三组肾脏组织形态学变化,收集血清检测肾功能变化,从氧化应激角度观察姜黄素的保护作用机制。结果:缺血再灌注模型组肾小管有明显形态改变,姜黄素处理后肾组织损害程度减轻。缺血再灌注组Paller评分显著高于对照组。经姜黄素处理后,与缺血再灌注组比较,Paller评分显著下降,差异有统计学意义(P<0.05)。缺血再灌注模型组BUN、Cr含量显著高于对照组。经姜黄素处理后,血清BUN、Cr含量均显著下降,差异均有统计学意义(P<0.05)。缺血再灌注模型组SOD含量显著低于对照组,MDA含量显著高于对照组。经姜黄素处理后,与缺血再灌注模型组比较,SOD含量显著升高,MDA含量显著下降(P<0.05)。结论:姜黄素可减轻缺血再灌注大鼠肾脏损伤程度,改善大鼠肾功能。姜黄素对大鼠缺血再灌注肾损伤的保护作用与提高大鼠血清SOD含量,降低MDA含量有关。
Objective:To observe the protective effect and mechanism of curcumin on renal ischemia-reperfusion injury in rats.Methods:45 male Sprague-Dawley rats were randomly divided into controlled group,ischemia-reperfusion model group and curcumin pretreatment group.The morphological changes of renal tissues were compared.The serum was collected for renal function changes and turmeric was observed from the perspective of oxidative stress.Results:There were no obvious morphological changes in the glomeruli and renal tubules of the controlled group.In the ischemia-reperfusion model group,the renal tubular epithelial cells showed edema and degeneration,and some cells showed vacuolar degeneration,brush-like necrosis and shedding.The renal tubules were narrowed,and tube-shaped and necrotic exfoliated cells could be seen.A small amount of renal tubules had powdered serum,proteinous fluid,and cell tube type.The interstitial was obviously hyperemia,edema,inflammatory cell infiltration,and perivascular vasodilation.The degree of renal tissue damage was reduced in the curcumin treatment group.The Paller score was significantly higher in the ischemia-reperfusion group than in the controlled group.After treatment with curcumin,the Paller score was significantly lower than that in the ischemia-reperfusion group,and the difference was statistically significant(P < 0.05).The levels of BUN and Cr in the ischemia-reperfusion model group were significantly higher than those in the controlled group.After treatment with curcumin,serum BUN and Cr levels were significantly decreased(P < 0.05).The SOD content in the ischemia-reperfusion model group was significantly lower than that in the controlled group,and the MDA content was significantly higher than that in the controlled group.After treatment with curcumin,the SOD content was significantly increased and the MDA content was significantly decreased compared with the ischemia-reperfusion model group(P < 0.05).Conclusion:Curcumin can alleviate the degree of renal injury in rats with ischemia-reperfusion and improve renal function.The protective mechanism may be related to the increase of serum SOD content and MDA content in rats.
