摘要
RNA病毒的反向遗传学是采用病毒的遗传材料(感染性克隆),在培养细胞或易感宿主中重新拯救出活病毒或类似病毒物质的过程.该技术在麻疹病毒属病毒中研究应用较多,尤以牛瘟病毒的研究开创了消灭麻疹病毒属病毒的先例,小反刍兽疫病毒和犬瘟热病毒的研究与应用还在进一步完善中.本文综述了反向遗传学技术在麻疹病毒属病毒基因结构与功能、致病机制、感染性克隆构建及新型疫苗研制等方面的研究进展.
Along with the development of reverse genetics technology at home and abroad,virus rescue has become possible.This method has been widely used for the study of measles virus.For RNA viruses,recombinant viruses have the characterized of genetic stability,strong infectivity,and no virulence return.Among them,the research on rinder-pest virus has set a precedent for the elimination of morbillivirus,which made measles virus under good control.Peste-des-petits-ruminants virus and canine distemper virus are still in the process of further improvement.Here,this paper discusses the pathogenesis of the virus,the structure of the gene,the construction of the vector and the novel vaccine by using the reverse genetics technology of the measles virus.
