白玉菇多糖对免疫抑制型小鼠的免疫调节作用 被引量:11

摘要 目的:研究白玉菇多糖对免疫抑制型小鼠脾脏、胸腺的免疫调节作用。方法:经超声波辅助热水浸提及Sevage法去蛋白得到白玉菇精多糖(RPHM)。将小鼠随机分为空白对照组、模型对照组、阳性对照组、RPHM低剂量组(30 mg/kg bw/d)、RPHM中剂量组(60 mg/kg bw/d)、RPHM高剂量组(120 mg/kg bw/d),每天同时间段进行灌胃,灌胃15 d,建立环磷酰胺(CPA)免疫抑制型小鼠模型。取小鼠脾脏、胸腺,测定其体重、脏器指数;血清细胞因子、单核-巨噬细胞吞噬能力;外周白细胞、红细胞和血小板计数并观察脾脏、胸腺的形态学。结果:与模型组相比,RPHM中剂量组(60 mg/kg bw/d)、高剂量组(120 mg/kg bw/d)可明显提升免疫抑制型小鼠的毛色、活动情况、身体状况、食欲、体重、脾脏指数和胸腺指数、单核-巨噬细胞吞噬功能、骨髓造血功能等指标;也可在一定程度上提高免疫抑制型小鼠IL-2、IL-12、IFN-γ和TNF-α的水平,并改善IL-6的分泌异常情况;还可明显改善免疫抑制型小鼠脾脏、胸腺的病变,改善结构完整度。结论:RPHM对免疫抑制型小鼠的脾脏和胸腺有明显的免疫调节作用。 Objective:To study the immunomodulatory effect of white Hypsizygus marmoreus on spleen and thymus of immunosuppressed mice.Methods:Ultrasonic assisted hot water extraction and sevage method to remove protein and obtain white polysaccharides.The mice were randomly divided into a blank control group,a model control group,a positive control group,a low dose group of RPHM(30 mg/kg bw/d),a medium dose group of RPHM(60 mg/kg bw/d),and a high dose group of RPHM.(120 mg/kg bw/d),intragastric administration at the same time every day to establish an immunosuppressive mouse model,15 days of gavage.The spleen and thymus of the mice were taken,and their body weight and organ index were measured.Serum cytokines and monocyte-macrophage phagocytosis ability;peripheral leukocytes,red blood cells and platelets were counted and the morphology of spleen and thymus were observed.Results:Compared with the model group the middle and high dose groups of RPHM,the hair color,activity,body condition,appetite,body weight,spleen index and thymus index,mononuclear-macrophage phagocytosis and bone marrow hematopoietic function of immunosuppressed mice were significantly improved.It can also increase the levels of IL-2,IL-12,IFN-γand TNF-αin immunosuppressive mice to acertain extent,and improve the abnormal secretion of IL-6;it can also significantly improve the spleen of immunosuppressed mice,thymic lesions,improve structural integrity.Conclusion:RPHM has obvious protective effects on spleen and thymus of immunosuppressed mice.
