摘要
目的探讨低浓度二甲基亚砜(Dimethyl sulfoxide,DMSO)对斑马鱼胚胎发育和成鱼生殖的影响。方法用体积比为2×10^-7%、2×10^-6%、2×10^-5%、2×10^-4%的二甲基亚砜处理斑马鱼受精卵96 h,成鱼96 h或10 d。将胚胎及成鱼交配所产受精卵24 hpf存活率,48 hpf及72 hpf孵化率与正常饲养的斑马鱼进行比较。结果2×10^-7%DMSO,2×10^-6%DMSO,和2×10^-5%DMSO处理胚胎后,24 hpf存活率,48 hpf和72 hpf孵化率都显著低于对照组(P<0.05)。二甲基亚砜处理成鱼96 h后,受精卵48 hpf孵化率都极其显著低于对照组(P<0.001)。二甲基亚砜处理成鱼10 d后受精卵72 hpf孵化率均显著高于对照组(P<0.05)。结论与正常水饲养相比,低浓度二甲基亚砜对斑马鱼胚胎发育以及成鱼生殖均有影响,建议使用二甲基亚砜作为助溶剂时应做阴性对照组。
Objective To evaluate the effect of low concentration dimethyl sulfoxide(DMSO)on the embryonic development and reproduction of zebrafish.Methods The zebrafish embryos were randomly distributed into 4 groups and were exposed to DMSO at concentrations of 2×10^-7%,2×10^-6%,2×10^-5%,and 2×10^-4%(v/v)for 96 hours.The survival rate at 24 hour post-fertilization(hpf)and the hatching rate at 48 hpf and 72 hpf were recorded.The adult zebrafish were exposed to DMSO at concentrations of 2×10^-7%,2×10^-6%,2×10^-5%,and 2×10^-4%(v/v)for 96 hours or 10 days.The survival rate at 24 hpf and hatching rate at 48 hpf and 72 hpf of the next generation were compared with the embryos and adult zebrafishes in normal water.Results In 2×10^-7%DMSO,2×10^-6%DMSO,and 2×10^-5%DMSO treatment groups,the survival rate at 24 hpf and hatching rate at 48 hpf and 72 hpf of embryos significant decreased compared with the control group(P<0.05).For the adult zebrafish exposed to DMSO for 96 h,the hatching rate of the next generation at 48 hpf significantly decreased compared with the control adult zebrafish(P<0.001).For the adult zebrafish exposed to DMSO for 10 days,the hatching rate at 72 hpf significantly increased compared with the control group(P<0.05).Conclusion Low concentration dimethyl sulfoxide has effect on Zebrafish embryos and adults zebrafish.
