四季秋海棠绿叶品系“超级奥林匹克”的离体再生体系建立 被引量:1

摘要 本研究以四季秋海棠绿叶品系“超级奥林匹克”的第3~5片完全展开叶片作外植体,从消毒条件、启动培养、增殖培养、生根培养等方面建立离体再生体系。结果为,(1)最佳消毒条件为0.25%NaClO处理15 min;(2)最佳启动培养基为MS+0.5 mg·L^-16-BA+0.12 mg·L^-1 NAA,分化率达到80%,且最早生芽时间为24 d;(3)最佳增殖培养基为MS+1.0 mg·L^-16-BA+0.1 mg·L^-1 NAA,增殖系数4.8;(4)最佳生根培养基为1/2 MS+0.1 mg·L^-1 NAA,生根率95%。(5)炼苗与移栽的方法为室温下炼苗一天后,选择珍珠岩∶腐殖质=1∶2作为移栽基质进行移栽,移栽成活率为96%。 Using the 3rd-5th fully developed leaves of Begonia semperflorens‘SuperOlympic’as explant,the regeneration system,including selection of disinfection,differentiation,multiplication and rooting,was established in this paper.Results showed that i)the best method of sterilization is treating leaf squares with 0.25%NaClO for 15 min,and the survival rate of explant could achieve 55%;ii)the best suitable differentiation medium is MS+0.5 mg·L^-16-BA+0.12 mg·L^-1 NAA,which made the shoot buds differentiation rate at 98%,and leaf squares producing buds on 24th day;iii)the best suitable multiplication medium is MS+1.0 mg·L^-16-BA+0.1 mg·L^-1 NAA which produced a coefficient multiplication of 4.8;iv)the best suitable rooting medium is 1/2 MS+0.1 mg·L^-1 NAA which made a rooting rate of 95%;v)after adaptation for 1 day,a mixture of perlite and humus(1:2,v/v)was selected as medium to transplant the tissue culture seedlings,and the survival rate could achieve 96%.
