摘要
我国是畜牧业大国,对于猪肉的食用量日益增加,但猪皮的利用率较低,造成了猪皮资源浪费。着眼于提高猪皮利用率及猪皮中胶原蛋白的提取率,通过酸水解工艺对胶原蛋白进行提取。分析不同盐析时间、不同酸处理pH及不同脱脂液浓度对胶原蛋白得率的影响,通过单因素试验确定进行正交试验的因素及水平;正交试验分析确定胶原蛋白提取的最佳条件为:盐析时间1.5h,酸处理pH 3.5,脱脂液浓度5%,此时,胶原蛋白提取量最高,为2.16g,得率为43.2%;根据极差分析可知,三个因素对胶原蛋白提取率的影响大小依次为盐析时间>pH>脱脂液浓度。同时,利用羟脯氨酸法测定提取物中胶原蛋白纯度,吸光度为0.154,胶原蛋白纯度为85.44%。
China is a big animal husbandry country, and the consumption of pork is increasing day by day, but the utilization rate of pigskin is low, which results in the waste of pigskin resources. The purpose of this study was to improve the utilization rate of pigskin and the extraction rate of collagen from pigskin by acid hydrolysis process. The effects of different salting out time, different acid treatment pH and different degreasing solution concentration on the yield of collagen were studied. The factors and level of orthogonal test were determined by single factor test. The optimum conditions of collagen extraction were determined by orthogonal test: Salting out time is 1.5 h, pH value is 3.5, degreasing liquid concentration is 5%.At this time, the extraction amount of collagen was 2.16 g, and the yield was 43.2%. According to range analysis, the effects of the three factors on the extraction rate of collagen were successively salting out time>pH>degreasing solution concentration.The collagen purity of the sample was determined by hydroxyproline method, the absorbance was 0.154 and the purity of collagen was 85.44%.
