鼻咽癌组织JAK2、STAT3表达变化及其临床意义 被引量:4

Expression changes of JAK2 and STAT3 in nasopharyngeal carcinoma and its clinical significance

摘要 目的观察鼻咽癌组织Janus蛋白酪氨酸激酶2(JAK2)、信号转导和转录激活因子3(STAT3)表达变化,并分析二者表达与患者临床病理特征和预后的关系。方法选择鼻咽癌患者85例,取鼻咽癌组织和癌旁正常组织,采用RT-qPCR法检测JAK2、STAT3 mRNA表达。比较鼻咽癌组织与癌旁正常组织JAK2、STAT3 mRNA相对表达量,分析二者表达与患者临床病理特征和预后的关系。结果鼻咽癌组织JAK2、STAT3 mRNA相对表达量均明显高于癌旁正常组织(P均<0.05)。Pearson线性相关分析显示,鼻咽癌组织JAK2 mRNA相对表达量与STAT3 mRNA相对表达量呈正相关关系(r=0.622,P<0.01)。鼻咽癌组织JAK2、STAT3 mRNA相对表达量均与TNM分期有关(P均<0.05),而与性别、年龄、吸烟史、组织分化程度、淋巴结转移无关(P均>0.05)。分别以JAK2、STAT3 mRNA相对表达量的均数为临界值,将鼻咽癌患者分为JAK2 mRNA高表达者43例与低表达者42例,STAT3 mRNA高表达者41例与低表达者44例。JAK2 mRNA高表达者与低表达者3年总体生存率分别为51.2%(21/43)、83.3%(35/42),STAT3 mRNA高表达者与低表达者3年总体生存率分别为48.7%(20/41)、79.5%(35/44)。JAK2、STAT3 mRNA高表达者3年总体生存率均明显低于其低表达者(χ^2分别为3.874、4.916,P均<0.05)。结论鼻咽癌组织JAK2、STAT3高表达,其表达变化与肿瘤TNM分期和患者预后有关。JAK2、STAT3有可能成为诊断鼻咽癌的肿瘤标志物和治疗靶标。 Objective To observe the expression changes of Janus protein tyrosine kinase 2(JAK2),signal transduction and transcriptional activator 3(STAT3)in the nasopharyngeal carcinoma tissues,and to analyze the relationship between the expression of JAK2,STAT3 and the clinicopathological characteristics and prognosis of patients.Methods Eighty-five patients with nasopharyngeal carcinoma were selected and the nasopharyngeal carcinoma tissues and adjacent normal nasal mucosa tissues were collected.The expression levels of JAK2 and STAT3 mRNA were detected by RT-qPCR.The relative expression levels of JAK2 and STAT3 mRNA between nasopharyngeal carcinoma tissues and adjacent normal nasal mucosa tissues were compared,and the relationship between the expression of JAK2 and STAT3 mRNA and the clinicopathological characteristics and prognosis of patients was analyzed.Results The relative expression levels of JAK2 and STAT3 mRNA in the nasopharyngeal carcinoma tissues were significantly higher than those in the adjacent normal nasal mucosa tissues(P<0.05).Pearson linear correlation analysis showed that the relative expression of JAK2 mRNA in nasopharyngeal carcinoma tissues was positively correlated with the relative expression of STAT3 mRNA(r=0.622,P<0.01).The relative expression of JAK2 and STAT3 mRNA in the nasopharyngeal carcinoma tissues was related to TNM stage(P<0.05),but not to sex,age,smoking history,degree of tissue differentiation,or lymph node metastasis(all P>0.05).The mean relative expression of JAK2 and STAT3 mRNA was taken as the critical value,and the nasopharyngeal carcinoma patients were divided into 43 patients with high JAK2 mRNA expression and 42 patients with low JAK2 mRNA expression,and 41 patients with high STAT3 mRNA expression and 44 patients with low STAT3 mRNA expression.The 3-year overall survival rates were 51.2%(21/43)and 83.3%(35/42)for patients with high and low JAK2 mRNA expression,and 48.7%(20/41)and 79.5%(35/44)for those with high and low STAT3 mRNA expression.The 3-year overall survival rate of the subjects with high expression of JAK2 and STAT3 mRNA was significantly lower than that of the subjects with low expression(χ^2=3.874 and 4.916,respectively,all P<0.05).Conclusion JAK2 and STAT3 are highly expressed in the nasopharyngeal carcinoma tissues,the high expression of JAK2 and STAT3 is related to TNM stage and poor prognosis,and both of them may be tumor markers and therapeutic targets for nasopharyngeal carcinoma.
