摘要
灰葡萄孢BcPDR1基因在病菌生长发育和致病过程中发挥重要作用,但其具体的分子机制尚未明确。本研究利用数字基因表达谱技术(Digital Gene Expression Profiling, DGE),获得了933个受BcPDR1基因影响的差异表达基因,GO富集分析发现差异表达基因主要集中在细胞过程、新陈代谢过程、细胞组分和催化作用等方面。利用数字基因表达谱数据,获得了14个差异表达的胞壁降解酶基因;热图分析和Real-time PCR分析发现,14个胞壁降解酶基因在突变体BCt89和ΔBcPDR1中的表达水平被明显上调或下调,确定了BcPDR1基因对胞壁降解酶基因的正负调控作用。试验结果为进一步阐明BcPDR1调控灰葡萄孢致病力的分子机制奠定了基础。
Botrytis cinerea BcPDR1 gene plays an important role in the growth,development and pathogenicity,but the molecular mechanism of its role remains to be explored.In this study,digital gene expression pro?ling was used to screen differentially expressed genes regulated by the BcPDR1 gene.933 differentially expressed genes that may be regulated by BcPDR1 gene were obtained.GO enrichment analysis showed that the differentially expressed genes were mainly related to cellular process,metabolic process,cell component,and catalytic function in B.cinerea.Using digital gene expression pro?ling data,14 differentially expressed cell wall degrading enzyme genes were obtained.Heat map and real-time PCR analysis revealed that 14 differentially expressed cell wall degrading enzyme genes were obviously upregulated or downregulated in mutant BCt89 andΔBcPDR1.It was determined that BcPDR1 positively or negatively regulated the expression of cell wall degrading enzyme genes.These results laid a foundation for further elucidating the molecular mechanism of BcPDR1 regulating the growth,development and pathogenicity of B.cinerea.
