副干酪乳杆菌对腹泻小鼠血清Th17细胞因子及其免疫相关激素水平的影响 被引量:4

Effect of Lactobacillus paracasei on serum Th17 cytokines and immune-related hormone levels in mice with diarrhea

摘要 为了研究副干酪乳杆菌类坚韧亚种(Lactobacillus paracasei subsp.Tolerans)对致病性大肠杆菌腹泻小鼠免疫机能及免疫相关激素水平的影响,试验将小鼠随机分为7组,分别为副干酪乳杆菌高、中、低剂量组,Nisin组,抗生素组,阴性对照组和正常对照组。采用碳廓血清法检测巨噬细胞的吞噬能力;ELISA法检测小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6 (interleukin-6,IL-6)、转化生长因子-β(transforming growth factor-β,TGF-β)、IL-17、IL-23和IL-1β以及内分泌激素糖皮质激素(glucocorticoid,GC)、三碘甲状腺原氨酸(triiodothyronine,T3)、甲状腺素(thyroxine,T4)、促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)和生长激素(growth hormone,GH)水平。结果表明:与阴性对照组相比,灌胃高剂量副干酪乳杆菌可显著提高腹泻小鼠单核巨噬细胞的吞噬能力(P<0.05);可极显著提高TGF-β水平(P<0.01),极显著降低IL-6和IL-1β水平(P<0.01),显著降低TNF-α、IL-17和IL-23水平(P<0.05);可显著降低GC含量(P<0.05),极显著降低ACTH含量(P<0.01),显著提高T3、T4和GH含量(P<0.05)。说明灌胃高剂量副干酪乳杆菌可以提高小鼠免疫力,调节内分泌激素水平,从而维持内环境的相对稳定,使机体处于健康状态。 This study is to investigate the effect of Lactobacillus paracasei subsp.Tolerans on the immune function and immune-related hormone levels in mice with diarrhea caused by enteropathogenic Escherichia coli.In this study,SPF KM mice were randomly divided into seven groups:high-,medium-and low-dose Lactobacillus paracasei(L.aracasei)groups,Nisin group,antibiotic group,negative control group,and normal control group.Macrophage phagocytosis was determined by using carbon particle clearance.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),transforming growth factor-β(TGF-β),IL-17,IL-23,IL-1β,glucocorticoid(GC),triiodothyronine(T3),thy roxine(T4),adrenocorticotropic hormone(ACTH)and growth hormone(GH)in serum of mice were detected by ELISA.The results showed that the phagocytic ability of mononuclear macrophages in diarrhea mice was significantly increased,compared with the negative control group(P<0.05);the level of TGF-βwas significantly increased(P<0.01).The levels of IL-1βand IL-6 were significantly decreased(P<0.01),the levels of TNF-α,IL-17 and IL-23 were significantly decreased(P<0.05);the content of GC was significantly decreased(P<0.05),and the content of ACTH was significantly decreased(P<0.01),the contents of T3,T4 and GH were significantly increased(P<0.05).It indicated that high dose of L.aracasei could increase the immunity of mice and regulate endocrine hormones,maintaining homeostasis to keep the body in good health.
