摘要
目的探讨加巴喷丁(gabapentin,GBP)在脊髓水平对带状疱疹后神经痛(postherpetic neuralgia,PHN)大鼠模型痛觉的抑制作用。方法通过对老龄大鼠足底皮下注射水痘-带状疱疹病毒(varicella zoster virus,VZV)建立PHN大鼠模型;并采用鞘内给药方法观察10、15、20μg三个剂量的GBP对该大鼠模型的长时程自发痛和机械性痛敏的抑制效应。结果大鼠模型表现出明显的机械性痛敏而无明显的热痛敏,能较好地模拟PHN临床表现。GBP对PHN大鼠模型长时程自发痛发挥抑制作用并呈剂量依赖性。GBP对PHN大鼠模型机械性痛敏无明显的抑制作用。结论 GBP可抑制PHN长时程自发痛并呈剂量依赖关系,GBP在脊髓水平对PHN能发挥镇痛作用。
Objective To observe whether gabapentin(GBP)has analgesic actions on the rat model of postherpetic neuralgia at the spinal level.Methods The rat model of postherpetic neuralgia(PHN)was built by intraplantar subcutaneous injection of varicella zoster virus(VZV)in aged rats.The analgesia action under 10,15 and 20μg GBP on the rat model of PHN were studied respectively by using intrathecal drug administration technique.Results In this model,mechanical hyperalgesia were produced obviously instead of the primary heat hyperalgesia,so that the rat model of PHN could mimic the chronic pain states that occured in PHN patients.Conclusion GBP can suppress the persistent spontaneous nociception responses of PHN in a dosedependent manner,and it may exert its analgesia action partially at the spinal level.
