芒针对骨关节炎大鼠炎症反应及镇痛作用影响的机制研究 被引量:4

摘要 目的:探究芒针对骨关节炎大鼠炎症反应及镇痛作用影响的作用机制。方法:将30只大鼠随机分为正常组、模型组及治疗组,每组10只。模型组与治疗组大鼠采用木瓜蛋白酶注射造模法制造骨关节炎模型,治疗组造模成功后给予双后肢“膝前”“后三里”芒针灸治疗,正常组及模型组不予干预。HE染色观察各组大鼠膝关节软骨组织病理变化;痛行为学方法检测大鼠双侧后肢热撤足阈值与机械性撤足阈值;采用还原酶法测定各组大鼠血清中NO含量、羟胺法测定SOD活力、硫代巴比妥酸法测定MDA含量;Western Blot、ELISA实验用于检测各组大鼠膝关节内细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)与诱导型一氧化氮合酶(iNOS)等蛋白表达变化。结果:HE染色结果显示,芒针治疗组有效缓解大鼠软骨组织病理变化;治疗后治疗组大鼠热痛阈值及机械性撤足阈值均较模型组显著提高(P<0.05);治疗组大鼠血清NO、MDA含量较模型组降低,而体内SOD含量则较模型组显著升高(P<0.05);WB及ELISA结果显示,治疗组大鼠膝关节软骨组织中IL-6、TNF-α及iNOS蛋白表达显著低于模型组(P<0.05)。结论:芒针可通过促进大鼠体内抗自由基损伤作用抑制大鼠炎症因子表达,提高OA大鼠镇痛阈值,缓解关节损伤。 Objective:To investigate the effect of elongated needle acupuncture on inflammation reaction and its analgesia effect in rats with osteoarthritis(OA).Methods:30 healthy SD rats were randomized divided into the normal group,the model group and the treatment group,with 10 rats in each group.OA model was established by injecting papain in the rats.The treatment group was given elongated needle acupuncture on points of“Xiqian”and“Housanli”after successful modeling,whereas the other two groups were not intervened.Pathological changes of knee articular artilage were observed by HE staining method.The pain behavior test was used to detect the mechanical and thermal withdraw threshold of the rats’bilateral hind paws.NO content was determined with nitrate reductase method,SOD activity was detected by hydroxylamine method,and thiobarbituric acid(TBA)method was used to determine MDA content.The expressions of IL-6,TNF-αand iNOS were measured by Western Blot and ELISA methods.Results:The pathologic changes of articular cartilage tissues were alleviated by elongated needle acupuncture.The mechanical and thermal withdraw threshold were significantly increased after the treatment(P<0.05).The serum contents of NO and MDA were significantly lowered,and SOD content was greatly increased in the treatment group than those in the model group(P<0.05).The expressions of IL-6,TNF-αand iNOS protein in the articular artilage were significantly decreased in the treatment group(P<0.05).Conclusion:Elongated needle acupuncture can inhibit the expressions of inflammatory cytokines,promote anti-free radical damage and increase the analgesic threshold in OA rats.
