利用EST-SSR检测阔短和狭长被片型美洲莲的遗传变异 被引量:1

摘要 为检测阔短被形(花被片形状)M512和狭长被形DP23两株美洲莲的基因组是否存在差异,在比较形态特征的基础上,对两个被形差异植株筛选多态性EST-SSR标记,并分析其生物学信息。在M512和DP23之间,20个形态指标中差异最为直观和显著的为被形(花被片形态及宽长比)、花径、花柄高度及粗度等花器官相关性状;217个EST-SSR标记中26对引物的PCR产物表现出条带多态性,其所对应核苷酸序列(基因)涉及器官发育、激素代谢、信号转导等多个方面。这表明阔短被形和狭长被形美洲莲在基因组水平存在差异,可进一步展开全基因组重测序等分析。 For detecting the genomic DNA differences between two types of American lotus,M512 with broad tepals and DP23 with narrow ones,the polymorphic EST-SSR markers were applied to investigate biological information based on their morphological characteristics.Among the 20 morphological indexes,significant differences between M512 and DP23 occurred in floral traits were tepal shape(the ratio of breadth to length),flower diameter,peduncle height and diameter.The 217 EST-SSRs used,26 EST-SSR primer pairs produced polymorphic bands between the two types of American lotus.The nucleotide sequences or genes obtained from those bands were associated with organ development,hormone metabolism,signal transduction,etc.It indicated that differences in genome level between M512 and DP23,could be further studied through genome-wide re-sequencing.
