赶黄草水提物对高脂饮食联合STZ诱导的Ⅱ型糖尿病大鼠的降血糖作用 被引量:14

摘要 研究了赶黄草水提取物对高脂饮食联合链脲佐菌素(STZ)诱导的Ⅱ型糖尿病大鼠的降血糖作用。随机选取10只SD大鼠作为正常对照组(NC),其余大鼠经4 w高脂饮食喂养后,腹腔注射小剂量STZ(35 mg/kg)建立Ⅱ型糖尿病大鼠模型。造模成功后随机分为:模型组(MC)、二甲双胍阳性对照组(PC,500 mg/kg/d)、赶黄草水提物组(PCP,545 mg/kg/d),每组10只。干预4 w后,检测各组大鼠血糖、血脂、抗氧化水平,肝、胰HE染色观察组织病变。结果表明,与模型组相比,赶黄草水提物组空腹血糖(FBG)、糖化血红蛋白(Hb A1c)水平分别降低43.93%、77.05%(p<0.05),血清高密度脂蛋白胆固醇(HDL-C)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)分别升高80.00%、76.32%、42.75%、32.12%(p<0.05),丙二醛(MDA)含量降低26.89%(p<0.01)。赶黄草水提物能显著降低Ⅱ型糖尿病大鼠血糖水平,改善胰岛素抵抗,其机制可能与其发挥抗氧化作用,减少氧化应激有关。 The hypoglycemic effect of extract of Penthorum chinense Pursh(PCP) on a high fat diet and streptozotocin(STZ)-induced diabetic rats was investigated. The efficacy of orally administer PCP(545 mg/kg/day) for 4 weeks were evaluated in high-fat diet and STZ(35 mg/kg.) induced diabetic rats, and metformin(500 mg/kg/day) was used as a positive control. A range of parameters, including blood glucose and lipid were tested to evaluate its anti-hyperglycemic effects. Moreover, superoxide dismutase(SOD), malonaldehyde(MDA), glutathione peroxidase(GSH-Px), and catalase(CAT) were performed to determinate their antioxidant activities. The fasting blood glucose and Hb Ac1 level of PCP treatment group decreased by 43.93% and 77.05%, respectively, when compared with the diabetic rat group(p<0.05). The activities of antioxidant enzymes including GSH-Px, SOD and CAT were significantly improved by 76.32%, 42.75% and 32.12%(p<0.05), respectively. PCP was also found to reduce the levels of MDA(p<0.01). Meanwhile, the histology results showed the extract of PCP could protect against liver and pancreas damages. This study showed that the extract of PCP modulated hyperglycemia in high-fat diet and STZ-induced diabetic rats, which might be attributed to enriching antioxidant enzymes activities.
