基于高通量转录组测序的白芷差异表达基因分析 被引量:7

摘要 【目的】为获得白芷转录组序列信息,了解不同产地间白芷的差异表达基因情况。【方法】以白芷根为研究对象,采用Illumina HiSeq X Ten平台对不同产地白芷进行高通量转录组测序,并对其差异表达基因进行筛选和分析。【结果】川白芷产地间、亳白芷产地间及川白芷与亳白芷间的差异基因总数分别为2686、1704、11549条,分别被GO富集注释11038、434、5604条,差异基因的主要功能与细胞组成、细胞、细胞器、结合、催化活性、细胞过程及代谢过程有关。KEGG通路分类显示,差异表达基因被分为4大类,24个小类,所在通路主要与代谢过程相关。与白芷主要化学成分香豆素和挥发油合成相关的4-香豆酸连接酶、丙氨酸氨解酶、3-羟基-3-甲基戊二酰辅酶A还原酶在川白芷中表达量升高,1-脱氧-D-木糖-5-磷酸合成酶、脱氢多萜醇焦磷酸合成酶在川白芷中表达量降低。【结论】通过转录组测序分析了不同产地白芷根基因差异表达状况,为白芷药材道地性品质的形成提供了理论依据。 【Objective】The present paper aimed to obtain the transcriptome dataset of roots of Angelica dahurica and understand the differentially expressed gene(DEG) of roots of Angelica dahurica in different areas. 【Method】The root transcriptome dataset of Angelica dahurica was obtained using the high-throughput sequencing platform Illumina HiSeq X Ten, followed by differentially expressed genes analyses.【Result】The number of differentially expressed gene between cbz, bbz and cbz vs bbz were 2686, 1704, and 11 549, respectively, and annotated by GO with 1713,1112, and 4418. And the functions of differentially expressed genes were related to cell composition, cells, organelles, binding, catalytic activity, cell processes, and metabolic processes. The KEGG pathway classification showed that the differentially expressed gene was divided into 4 categories and 24 subcategories, and the pathway was mainly related to the metabolic process.The expression of genes encoding 4-coumarate CoA ligase, Phenylalanine ammonia-lyase 3 and 3-hydroxy-3-methylglutaryl-coenzyme A reductase increased in cbz, while, the expression of genes encoding dehydrodolichyl diphosphate synthase and probable 1-deoxy-D-xylulose-5-phosphate synthase 2 decreased. 【Conclusion】In this study, the differentially expressed gene of Angelica dahurica root was analyzed by transcriptional group sequencing, which provided theoretical basis for the formation of quality of Angelica dahurica in the future.
