白花蛇舌草紫外指纹图谱共有峰率和变异峰率双指标序列分析法 被引量:2

摘要 【目的】建立云南不同产区白花蛇舌草的紫外指纹图谱,阐明不同产区间白花蛇舌草的相似程度和差异,为白花蛇舌草的真伪鉴定和品质鉴定评价奠定基础。【方法】采用紫外分光光度法测定云南23个不同产区白花蛇舌草紫外图谱,考察白花蛇舌草乙酸乙酯、甲醇、水三种极性溶剂的提取物各波段的稳定性及重现性,对紫外图谱进行平均值、平滑和二次微分处理,计算白花蛇舌草不同提取物的共有峰率和变异峰率。【结果】建立的白花蛇舌草指纹图谱:最佳称样量为0.5000 g,乙酸乙酯、甲醇和水3种溶剂最佳提取时间分别为30、40、30 min,3种溶剂提取液在48 h内稳定。白花蛇舌草紫外指纹图谱显示,不同产区样品间最大共有峰率为92.3%,最小共有峰率为22.2%。【结论】利用紫外光谱结合共有峰率和变异峰率双指标序列分析法能阐明不同产区间白花蛇舌草的的相似程度,为白花蛇舌草药材真伪鉴定和品质评价奠定基础。 【Objective】The ultraviolet fingerprints of Hedyotis diffusa Willd from twenty-three different areas in Yunnan were established.It also could be used to clarify the similarity and diversity level among the samples from different areas,which provided a foundation for authen-ticity identification and quality evaluation of Hedyotis diffusa Willd.【Method】The UV spectra of Hedyotis diffusa Willd from twenty-three different origins in Yunnan province were determined by ultraviolet and visible spectrophotometer.The dual indexes of common peak ratio and variant peak ratio,mean value,smoothness and second differential methods were analyzed by using the UV-spectrum data to investigate the stability and repeatability of various wave bands of three different polarity solvent extracts(ethyl acetate,methanol and water)of Hedyotis diffusa Willd.The common peak ratio and variant peak ratio for the different extracts of Hedyotis diffusa Willd were calculated.【Result】The optimal sample weight was 0.5000 g.The best extraction time of thyl acetate,methanol and water was 30,40,30 min respectively,and their extract was stable in 48 hours.The fingerprint spectral implicated that the maximum common peak rate was 92.3%,while the mini-mum was 22.2%.【Conclusion】The dual index sequential analysis could be used exactly in the qualitative evaluation of Hedyotis diffusa Willd,and clarified the similarity level among different areas,also applied the basement for the authenticity identification and quality evalua-tion of Hedyotis diffusa Willd.
