慢病毒介导的过表达载脂蛋白A1宫颈癌稳定细胞系建立的实验研究

摘要 目的采用慢病毒载体构建稳定过表达载脂蛋白A1(apolipoprotein A1,ApoA1)基因的SIHA、HELA细胞系,为研究ApoA1在宫颈癌中的作用机制奠定基础。方法构建过表达ApoA1基因的慢病毒载体,感染SIHA、HELA细胞并进行嘌呤霉素抗性筛选,构建稳定过表达ApoA1基因的SIHA、HELA细胞系,采用实时荧光定量PCR(RT-qPCR)和Western blot方法检测ApoA1 mRNA和蛋白的表达水平。结果RT-qPCR显示SIHA及HELA细胞转染组中ApoA1mRNA表达量较阴性对照组明显提高(P<0.01),Western blot提示ApoA1蛋白在SIHA及HELA细胞中低表达,转染组ApoA1蛋白水平的表达明显高于对照组。结论成功构建稳定过表达ApoA1基因的宫颈癌SIHA、HELA细胞系。
