摘要
为培育具有药用保健作用的胡萝卜品种提供参考,以黑田五寸胡萝卜为材料,克隆胡萝卜DcBMT基因,并分析其进化和表达情况。结果表明:克隆得到的胡萝卜DcBMT基因,其ORF框长为1080 bp,可编码359个氨基酸,该蛋白酶分子质量为39.25 kD,等电点pI为6.31,含有5个保守的Box结构和3个保守基序。胡萝卜和白芷的BMT氨基酸序列属同一分枝,而与其他伞形科植物存在一定距离。胡萝卜DcBMT基因在根发育3 d和60 d的表达量最高,在收获时(90 d)表达量最低。DcBMT基因在胡萝卜肉质根发育过程中均有表达,胡萝卜肉质根中可能有佛手酚及佛手柑内脂合成及物质的积累。
The evolution and expression of DcBMT gene cloned from Kuroda(a carrot variety)are analyzed to provide a reference for breeding carrot varieties with medicinal health care function.Result:The ORF of cloned carrot DcBMT gene is 1080 bp,encoding a protein of 359 amino acid.The protease has a molecular weight of 39.25 kD and isoelectric point pI of 6.31,and contains 5 conservative Box structure and 3 conservative motif.BMT amino acid sequence of carrot and Radix angelicae belongs to the same branch but there is a certain distance in BMT amino acid sequence between carrot and other umbelliferae plants.The expression of DcBMT gene in root is the highest at 3 d and 60 d and the lowest at harvesting stage(90 d).DcBMT gene is expressed during carrot fleshy root development.Carrot fleshy root may have synthesis of bergaptol and bergapten and matter accumulation.
