摘要
目的:研究负载甘精胰岛素PLGA(聚乳酸-羟基乙酸共聚物)微球种植体对2型糖尿病大鼠种植体周围成骨的影响。方法:设计并制作一种应用于大鼠等小型动物的种植体,根据种植体表面处理将实验分为甘精胰岛素组(n=5)和对照组(n=5)。甘精胰岛素组:种植体表面负载甘精胰岛素PLGA微球植入大鼠右侧膝关节内侧,对照组:植入空白微球负载的种植体。每周动态测量血糖值及ISQ值种植体稳定系数(implant stability quotient,ISQ)。种植体植入第6周后拍摄CT并计算种植体骨结合率。结果:两组动态血糖值均高于18mmol/L,且均不具有统计学差异(P>2.61)。而甘精胰岛素组第4、5、6周ISQ值及第6周种植体骨结合率显著高于对照组(P<0.001)。结论:设计制作的光滑表面短种植体能够成功应用于GK大鼠,获得良好的生物相容性,能够动态测量ISQ值,在高血糖环境下获得一定的骨结合,PLGA微球负载的甘精胰岛素对2型糖尿病大鼠种植体周围成骨具有促进作用。
Objective:To evaluate the effect of insulin-loaded PLGA(polylactic-co-glycolic acid)microspheres on bone healing around implants in GK rats(type 2 diabetic rats).Methods:A type of implant for small animals such as rats was designed and manufactured.According to the surface treatment of the implant,the animals were divided into the insulin glargine group(n=5)and the control group(n=5).Insulin glargine group:GK rats were implanted with insulin glargine PLGA microsphere in their interior of right knee joint;control group(n=5):GK rats were implanted with blank PLGA microsphere.Blood glucose level and ISQ were measured each week,and the osseointegration rates of implant were analyzed by CT scan 6 weeks after implantation.Results:The blood glucose level were both above 18mmol/L,and of no significant difference between the two groups(P>2.61).However,the level of ISQ in insulin glargine group were significant higher than that in the control group at the 4th,5th and 6th weeks(P<0.001).Furthermore,the implant osseointegration rate in insulin glargine group was significant higher than that in the control group at the 6th week(P<0.001).Conclusions:The self-designed short titanium implant was successfully applied to the GK rat with satisfied biocompatibility.The ISQ value can be dynamically measured.And the osseointegration was acceptable under high glucose condition.The self-desighed implants loaded with glargine PLGA microsphere promoted osteogenesis in GK rats.
