摘要
本试验旨在揭示西藏藏猪与大约克猪Mx1基因的表达差异。分别选择西藏林芝市6月龄藏猪与大约克猪各10头,采用荧光定量PCR方法检测Mx1基因在肝脏、脾脏中的相对表达量,采用ELISA方法测定2个品种猪血清中α干扰素水平。结果表明:Mx1基因在心脏、肝脏、脾脏、肺脏、肾脏、背最长肌、脑、下丘脑多个组织中均有表达;在肝脏、脾脏组织中藏猪的表达量极显著高于大约克猪(P<0.01),2个品种猪血清中,α干扰素水平与m RNA水平表达情况趋势一致,为藏猪极显著高于大约克猪(P<0.01)。由此结果推断,Mx1基因的表达及α干扰素水平在藏猪与大约克猪中存在明显差异,推测这可能与藏猪抵抗力较强存在密切相关,为进一步开展Mx1基因在藏猪抗病性能研究中提供重要依据。
The purpose of this study is to reveal the difference in Mx1 gene expression between Tibetan pigs and Yorkshire pigs.Ten Tibetan pigs and ten Yorkshire pigs at six months were selected,and the relative expression of Mx1 gene in the liver and spleen was detected by fluorescent quantitative PCR.The interferonαlevels in the serum of the two breeds of pigs were measured by ELISA.The results showed that Mx1 gene was expressed in many tissues of heart,liver,spleen,lung,kidney,longest dorsal muscle,brain,and hypothalamus,and the expression level of Tibetan pigs in liver and spleen tissues was significantly higher than Yorkshire pigs(P<0.01).The levels of interferon alpha and mRNA levels in the serum of the two breeds of pigs showed a consistent trend,in which both levels in Tibetan pigs was significantly higher than those in Yorkshire pigs(P<0.01).From the results,it can be inferred that the expression of Mx1 gene and the level of interferon alpha are significantly different between Tibetan pigs and Yorkshire pigs,which may be closely related to strong resistance of Tibetan pigs.In addition,this study provides an important basis for further research on disease resistance of Tibetan pigs.
