摘要
目的探讨USP39的表达水平对卵巢癌细胞放疗敏感性的影响。方法采用慢病毒转染敲降USP39,ES2细胞经或者不经6Gy X线照射后,采用流式细胞仪检测细胞凋亡情况,采用Western Blot检测凋亡相关蛋白及DNA损伤相关蛋白的表达水平。结果与未行X线照射组比较,ES2细胞经USP39敲降后行X线照射,细胞凋亡率、凋亡相关蛋白(cleaved-PARP,cleaved-Caspase3)及DNA损伤相关蛋白(γ-H2A.X)表达水平明显升高。结论USP39表达水平与卵巢癌的放疗敏感性密切相关。
Objective To explore the relationship between the USP39 gene expression and the change of radiosensitivity of human ovarian cancer cell.Methods Lentiviralvector mediated transfection was used to scilencing USP39.Under 6 Gy X ray radiation or not,the cell apoptosis of each group were detected by flow cytometry,the expression level of apoptosis and DNA damage associatied protein of ES2 cells was detected by Western Blot.Results The apoptosis rate and the expression level of apopotosis and DNA damage associatied protein of USP39 silencing ES2 cells was upregulated with X ray radiation(P<0.05).Conclusion The expression level of USP39 was associated with radiosensitivity.
