重度子痫前期胎盘组织中miR-210及GPD1L的相关性分析

摘要 目的分析miR-210及GPD1L在重度子痫前期胎盘组织中的表达及其相关性。方法选取2018年1月-2018年12月在笔者所在医院住院并行剖宫产分娩的40例重度子痫前期孕妇为试验组及40例产前检查正常并行剖宫产分娩的健康孕妇为对照组。采用免疫组织化学法检测两组胎盘组织中HIF-1α蛋白及GPD1L蛋白的表达情况。采用real-time PCR(RT-PCR)法分别检测两组胎盘组织中miR-210、GPD1L mRNA及HIF-1αmRNA的含量。结果 (1)与对照组相比,重度子痫前期组胎盘组织GPD1L蛋白的表达水平明显降低,其阳性表达率为(32.5%,P<0.05);HIF-1α蛋白的表达水平明显升高,其阳性表达率为(65%,P<0.05);重度子痫前期胎盘组织中miR-210的表达水平较高(10.010±3.153,t=6.942,P<0.001),GPD1LmRNA的表达水平较低(0.440±0.264,t=-5.013,P<0.001);HIF-1αmRNA的表达水平较高(7.331±2.407,t=6.397,P<0.001)。(2)在重度子痫前期胎盘组织中,miR-210的表达水平与GPD1L mRNA的表达水平存在明显的负相关(r=-0.911,P<0.001)。结论 miR-210可能通过抑制GPD1L mRNA的表达上调HIF-1α蛋白的含量而参与重度子痫前期的病理生理过程。 Objects The expression of miR-210 and GPD1L in the placenta of severe preeclampsia and their correlation was analyzed. Methods Forty gravidas who were admitted to the Third Affiliated Hospital of Zhengzhou University with sPE were enrolled in experimental group and an equal number of healthy gravidas who experienced uneventful pregnancy to those in the experimental group served as controls from January 2018 to December 2018.Levels of Hypoxia-induciblefactor-1α(HIF-1α)at protein and glycerol-3-phosphatedehydrogenase1 like(GPD1L) at protein in placenta tissues were detected by immunohistochemistry. Real-time PCR(RT-PCR)was used to detect the expression of miR-210、GPD1LmRNA and HIF-1αmRNA in placenta tissues of the two groups. Results(1)Compared to the control group,the sPE group showed decreased GPD1L at protein level in both placenta tissues and positive rate(32.5%,P<0.05),increased HIF-1α at protein level in both placenta tissues and positive rate(65%,P <0.05),increased expression of miR-210 in placenta tissues(10.010 ±3.153,t =6.942,P <0.001),decreased expression of GPD1L mRNA in placenta tissues(0.440 ±0.264,t =-5.013,P <0.001),increased expression of HIF-1αmRNA in placenta tissues(7.331±2.407,t=6.397,P<0.001).(2)Expression of miR-210 level in placenta tissues of the sPE group was negatively correlated with the level of GPD1L mRNA in placenta tissues(r =-0.911,P <0.001). Conclusions miR-210 may be involved in the pathophysiological process of severe preeclampsia by inhibiting the expression of GPD1L mRNA and up-regulating the HIF-1α protein concentration.
