二冬二母二草汤对慢性阻塞性肺疾病相关肺动脉高压平滑肌细胞增殖的影响及作用机制研究 被引量:6

摘要 目的:研究二冬二母二草汤对慢性阻塞性肺疾病(Chronic Obstructive Pulmonary Diseases,COPD)相关肺动脉高压大鼠模型的肺动脉平滑肌细胞(Pulmonary Arterial Smooth Muscle Cells,PASMCs)增殖的影响及作用机制。方法:体外培养PASMCs,分为对照组、模型组、二冬二母二草汤组(低、中、高浓度组)、Rho激酶抑制剂组。采用CCK-8方法测定PASMCs增殖水平,通过Western blot测定PASMCs中ROCK1、ROCK2表达水平。结果:与正常对照组相比,模型组的PASMCs增殖及ROCK1、ROCK2表达均明显升高(P<0.01);与模型组相比,低、中、高剂量组及Rho激酶抑制剂组的PASMCs增殖及ROCK1、ROCK2表达均明显降低(P<0.01或P<0.05),其中以高剂量组及Rho激酶抑制剂组降低显著。结论:二冬二母二草汤可明显抑制COPD相关肺动脉高压大鼠的PASMCs增殖,作用机制与调节Rho激酶信号通路有关。 Objective:To study the effect and action mechanism of Erdong-Ermu-Ercao(Erdong:asparagus and ophiopogon;Ermu:anemarrhenae and bulbus fritillariae thunbergii;Ercao:Houttuynia cordata and liquorice)Decoction(二冬二母二草汤)on pulmonary arterial smooth muscle cells(PASMCs)proliferation in COPD related pulmonary arterial hypertension rat models.Methods:PASMCs were cultured in vitro,and divided into Control Group,Model Group,Decoction Group(Low/Middle/High Concentration Group)and Rhokinase Inhibitor Group.CCK-8 was used to measure the proliferation level of PASMCs.Westernblot was applied to determine the level of ROCK1 and ROCK2 expression.Result:Compared with regular Control Group,the proliferation of PASMCs and level of ROCK1 and ROCK2 expression of Model Group were higher(P<0.01).Compared with Model Group,the proliferation of PASMCs and level of ROCK1 and ROCK2 of Decoction Group and Rho-kinase Inhibitor Group were lower(P<0.01 or P<0.05).The indexes of High Concentration Group and Rho-kinase Inhibitor Group were obviously lower.Conclusion:Erdong-Ermu-Ercao Decoction could inhibit the proliferation of PASMCs in COPD related pulmonary arterial hypertension rats.The action of mechanism had relationship with the signal of Rho-kinase.
