谷子OFP基因家族的鉴定及表达分析 被引量:3

摘要 谷子是我国重要的粮饲作物,具有抗旱耐贫瘠的特点,OFP家族是植物特有的一类调节植物生长和发育的转录因子,参与调控植物的抗逆性。为了探索谷子OFP家族基因的特征和功能,通过基因定位、基因结构分析、motif预测、进化分析与顺式作用元件分析及亚细胞定位预测来解析谷子OFP家族基因的特征,分析谷子与拟南芥和水稻基因组中的OFP家族基因的共线性、选择压力,最后分析谷子OFP家族基因的表达模式。结果表明,共筛选到谷子OFP家族基因34个,分布在8条染色体上;进化分析发现,谷子OFP家族基因可以分为3个亚类;motif预测中共鉴定到8个motif基序;顺式作用元件分析发现,与干旱响应相关的OFP基因共22个;与拟南芥的5个基因、水稻的31个基因存在共线性;18个OFP家族基因被预测定位到细胞核内;谷子OFP家族基因在穗中表达量最多,干旱胁迫下的OFP基因大多表现下调,具有时空特异性。OFP家族基因Seita.2G184300、Seita.7G093000、Seita.3G008900的表达差异可能是造成豫谷1号和安-04抗旱性差异较大的原因之一。结果可为谷子OFP家族基因的功能研究提供基础。 Foxtail millet is an important foodstuff crop in China.It has the characteristics of drought resistance and barrenness tolerance.The OFP family is a unique transcription factor regulating plant growth and development,and also participates in the regulation of plant stress resistance.This study will preliminarily explore the characteristics and functions of the OFP family genes in foxtail millet.The characteristics of OFP family genes in foxtail millet were analyzed by gene mapping,gene structure analysis,motif prediction,evolutionary analysis,cis-acting element analysis and subcellular localization prediction.At the same time,the collinearity and selective pressure analysis of OFP family genes in foxtail millet,Arabidopsis and rice genomes were analyzed.Finally,we analyzed the gene expression patterns of OFP family in foxtail millet.The results showed that there were thirty-four genes of foxtail millet OFP family were screened and distributed on eight chromosomes.Evolutionary analysis revealed that OFP family genes in foxtail millet could be divided into three subgroups.Eight motifs were identified in motif prediction.Cis-acting element analysis showed that twenty-two genes related to drought response,the foxtail millet OFP family gene was colinear with five Arabidopsis genes and 31 genes of rice,and eighteen OFP family genes were predicted to be located in the nucleus.The OFP family genes in foxtail millet were most expressed in panicles,and the OFP gene was more down-regulated under drought stress,which had temporal and spatial specificity.The difference of expression of Seita.2G184300,Seita.7G093000 and Seita.3G008900 may be the cause of the difference of drought resistance between Yugu 1 and An-04.This study will provide a basis for functional study of OFP family genes in foxtail millet.
