摘要
目的:探究舌鳞癌中HuR与端粒酶活性和端粒之间的关系。方法:收集6例舌鳞癌患者癌症组织及周围正常组织分别检测其端粒酶活性和HuR表达。在舌癌SCC25细胞中敲低HuR探究其对端粒酶和端粒的影响。结果:舌癌组织中端粒酶活性及HuR表达水平均比正常组织高(P<0.05);HuR能够与端粒酶TERC结合;在SCC25细胞中敲低HuR并不影响TERT和TERC的表达,但能够减弱两者的结合,进而降低端粒酶活性,同时长期敲低HuR使细胞端粒明显缩短。结论:在舌鳞癌中HuR通过结合在TERC上促进端粒酶组装进而调控端粒酶活性和端粒长度。
Objective:To investigate the relationship between RNA binding protein HuR(HuR)and the telomerase activity in tongue squamous carcinoma(TSC).Methods:The telomerase activity and the expression of HuR in TSC tissues and normal tissues collected from 6 cases were respectively detected.Furthermore,the effect of knockdown of HuR on telomerase expression and telomere length was tested in SCC25 cells.Results:The telomerase activity and HuR expression level were significantly higher in TSC than those in normal tissues(P<0.05).HuR was demonstrated to bind to TERC at cellular levels;HuR knockdown in SCC25 cells did not influence the expression level of TERC and TERT,but decreased the assembly of TERC and TERT,and negatively regulated the telomerase activity.Meanwhile,long term HuR knockdown of HuR decreased the length of telomere.Conclusion:HuR can positively regulate telomerase activity and telomere length through binding to the TERC,which enhances the assembly of telomerase in TSC.
