化浊解毒方对多氯联苯暴露大鼠胰岛素抵抗及糖脂代谢的影响 被引量:9

Effect of Huazhuo Jiedu Decoction on Insulin Resistance and Glucose and Lipid Metabolism in Rats Exposed to Polychlorinated Biphenyl

摘要 目的研究化浊解毒方对多氯联苯(polychorinated biphenyls, PCBs)暴露大鼠胰岛素抵抗及糖脂代谢的影响。方法雄性ZDF(fa/fa)大鼠,根据OGTT合格者随机分为模型组与PCBs染毒组,模型组6只,PCBs染毒组12只,另设6只雄性ZDF(fa/+)大鼠为正常对照组,正常组予普通饲料喂养+等体积玉米油灌胃,模型组予PMILabdiet 5008饲料+等体积玉米油灌胃,染毒组予PMILabdiet 5008+PCBs(5μg·kg^-1)灌胃染毒,共染毒4周,染毒后,将PCBs染毒组随机分为染毒组与治疗组(化浊解毒方治疗组),治疗组予化浊解毒颗粒按生药6 g·kg^-1·d^-1灌胃给药,余组灌服等体积的生理盐水,连续处理4周。对不同组别采用葡萄糖氧化酶法进行OGTT检测,放射免疫法测其空腹血清胰岛素水平,全自动生化分析仪检测大鼠血清的三酰甘油(TG)、胆固醇(TC)水平,比色法检测游离脂肪酸(FFA)水平。结果与染毒组比较,治疗组血糖、空腹胰岛素水平、胰岛素抵抗指数及血脂水平均下降,胰岛素敏感指数升高,差异具有统计学意义(P <0.05)。结论化浊解毒方具有显著改善PCBs暴露大鼠胰岛素抵抗及糖脂代谢的作用。 Objective To study the effects of Huazhuo Jiedu decoction on insulin resistance and glycolipid metabolism in rats exposed to polychlorinated biphenyls(PCBs). Methods Male ZDF(fa/fa) rats were randomly divided into model group and PCBs exposure group according to OGTT, 6 model groups, 12 PCBs exposure groups, and 6 male ZDF(fa/+) rats. For the normal control group, the normal group was fed with normal feed and equal volume of corn oil. The model group was given PMILabdiet5008 feed + equal volume corn oil, and the exposed group was given PMILabdiet5008+ PCBs(5 μg/kg). After 4 weeks of exposure, the exposed groups of PCBs were randomly divided into the exposure group and the treatment group(Huazhuo Jiedu decoction treatment group), and the treatment group was treated with Huazhuo Detoxification Granules according to the crude drug 6 g/kg·d. The remaining group was given an equal volume of physiological saline for 4 weeks. The glucose oxidase method was used to detect OGTT in different groups. The fasting serum insulin level was measured by radioimmunoassay. The serum triglyceride(TG) and cholesterol(TC) levels were detected by automatic biochemical analyzer. Colorimetric detection was used to test free fatty acid(FFA) levels. Results Compared with the exposure group, the blood glucose, fasting insulin level, insulin resistance index and blood lipid level decreased, and the insulin sensitivity index increased in the treatment group, the difference was statistically significant(P<0.05). Conclusions Huazhuo Jiedu decoction has a significant effect on improving insulin resistance and glycolipid metabolism in rats exposed to PCBs.
