8种易裂果实中果胶酶生物信息学分析 被引量:2

摘要 [目的]果胶酶是裂果中的一种关键酶,在植物中主要有多聚半乳糖醛酸酶和果胶酯酶2种,本研究旨在探究其理化性质,为进一步了解果胶酶生理调控机制以及裂果的发生机制奠定基础。[方法]采用生物信息学的方法,以拟南芥为对照,对番茄、苹果、白梨、葡萄、枣、甜樱桃、甜橙、桃8种不同易裂果物种中的2种果胶酶理化性质进行分析。[结果]在8种植物中,果胶酯酶大小为536~571 aa,多聚半乳糖醛酸酶大小为393~460 aa,它们大部分都定位在细胞膜并且有一段长度为22 aa的跨膜结构,等电点多碱性,都有多个糖基化位点,且果胶酯酶有4个高度保守基序,多聚半乳糖醛酸酶有2个高度保守的基序,在各自家族中,蛋白质结构上相似,进化关系保守。[结论]这些分析为裂果病变机理的研究及预防治理提供了理论基础,也为新品种的培育提供了新思路。 [Objective]Pectinases is a group of enzyme family involved in pectin degradation and fruit cracking producing,in which two major enzymes are polygalacturonase and pectinesterase existing in the plants.This study aimed to explore the physicochemical properties and provide scientific basis for further understanding of pectinase physiological regulation mechanism and fruit cracking mechanism.[Methods]In this study,the physicochemical properties of two pectinases in eight different species with fragile fruits,Arabidopsis thaliana,Lycopersicon esculentum,Malus domestica,Pyrus bretschneideri Rehd,Vitis vinifera,Ziziphus jujuba,Cerasus avium,Citrus sinensis,and Amygdalus persica,were analyzed by bioinformatics technique.[Results]Among eight species,pectinesterase size was ranged between 536 to 571 aa,and galacturonic acid enzyme polygalacturonase size was measured between 393 and 460 aa with majority located in the cell membrane and possessed a transmembrane structure at a length of 22 aa.The isoelectric point of two enzymes in most tested plants was alkaline with several glycosylation sites,and pectin esterases possessed four highly conservative motif.Galacturonic acid polymer enzyme owned two highly conservative motif and showed the protein structure similarity and conservative evolutionary relationships in the families they belong to.[Conclusion]These analyses provided a theoretical basis for the future study on the underlying mechanism of fruit cracking and the control strategy development,and provided new ideas for the cultivation of new varieties.
