Correlation between VISTA expression and prognosis of patients with pancreatic cancer

目的探讨T细胞活化的V结构域免疫球蛋白(VISTA)在胰腺癌组织中的表达情况,以及其与胰腺癌患者临床病理特征及预后的相关性,并探讨其作为胰腺癌一种潜在预后预测标记物的可能性。方法采用组织芯片进行免疫组化实验,检测73例胰腺癌组织及其对应的癌旁组织中VISTA的表达差异。采用χ^2检验或Fisher精确概率法分析VISTA表达水平与胰腺癌患者临床病理特征的相关性,采用Kaplan-Meier生存曲线、单因素Cox风险比例模型分析胰腺癌患者临床病理特征与预后的相关性。结果对组织芯片进行免疫组化分析发现,VISTA在胰腺癌组织中的表达量明显低于其对应的癌旁组织。以高表达和低表达分组时,VISTA的表达与淋巴结转移(P=0.030)、TNM分期(P=0.043)、病变位置(P=0.017)及总生存率(P=0.023)有关,而与患者性别、年龄、肿瘤直径、远处转移、病理分级无关(P均>0.05)。VISTA高表达与患者的良好预后显著相关,且无淋巴结转移及TNM分期较低的患者生存率更高。结论VISTA在胰腺癌组织中表达明显下调,且与患者的不良临床病理特征及预后相关。VISTA可作为一种潜在的胰腺癌预后预测标志物,为胰腺癌免疫治疗方法提供了一定的思路。

Objective To investigate the expression of V-domain immunoglobulin suppressor of T cell activation(VISTA)in pancreatic cancer and its correlation with clinicopathological features and prognosis of pancreatic cancer patients,and to explore the possibility of VISTA as a potential prognostic marker of pancreatic cancer.Methods Tissue microarray was used for immunohistochemistry to detect the difference of VISTA expression in 73 pancreatic cancer tissues and their corresponding adjacent tissues.χ^2 test or Fisher′s exact test were used to analyze the correlation between VISTA expression level and clinicopathological characteristics of pancreatic cancer patients.Kaplan-Meier survival curve,Cox proportional hazard model were used to analyze the correlation between clinicopathological characteristics and prognosis of pancreatic cancer patients.Results Immunohistochemical analysis of tissue microarray showed that the expression of VISTA in pancreatic cancer tissue was significantly lower than that in its corresponding paracancerous tissue.The expression of VISTA was related to lymph node metastasis(P=0.030),TNM stage(P=0.043),lesion location(P=0.017)and overall survival rate(P=0.023),but not related to the patient′s gender,age,tumor diameter,distant metastasis and pathological grade(P>0.05).Kaplan-Meier survival curve and Cox proportional hazard model suggested that the high expression of VISTA was significantly related to the good prognosis of patients,and the survival rate of patients without lymph node metastasis and with lower TNM stage was higher.Conclusions VISTA is significantly down-regulated in pancreatic cancer tissue,and it is related to the poor clinicopathological characteristics and prognosis of patients.It can be used as a potential prognostic marker of pancreatic cancer,which provides some ideas for immunotherapy for pancreatic cancer.
