阿尔茨海默病患者IL-1β、IL-6、Alpha-2-macroglobulin与认知障碍程度关系的研究 被引量:9

Study on the relationship between interleukin-1beta,interleukin-6,alpha2M and the degree of cognitive impairment in patients with Alzheimer's disease

摘要 目的探讨阿尔茨海默病(AD)患者血清IL-1β、IL-6和α2巨球蛋白(α2M)水平的变化及意义。方法对40例AD患者进行简易精神状态检查(MMSE)评分,用酶联免疫吸附法检测血清中IL-1β、IL-6和α2M水平,并与帕金森病(PD)患者及正常对照组进行比较。分析AD患者中各因子与MMSE评分的相关性,分析AD中α2M水平与其它各因子的相关性。结果①PD患者中IL-1β水平明显升高(P<0.05),AD患者IL-1β水平较正常对照组稍高、但差异无统计学意义(P>0.05);②AD、PD患者血清IL-6水平明显升高(P<0.05),两组间无差异(P>0.05);③AD、PD组α2M水平较对照组无明显变化(P>0.05);④AD中IL-6水平与MMSE评分呈负相关(r=-0.281,P=0.016)。AD中α2M水平与IL-1β、IL-6无相关性(P>0.05)。结论AD、PD的发病过程伴随着外周炎症反应,IL-6有可能与AD认知功能障碍程度有关,IL-1β与AD认知障碍程度无关,α2M尚不能作为AD的诊断标志物。 Objective To investigate the alterations of interleukin-1-beta(IL-1β),interleukin-6(IL-6)and alpha2macroglobulin(α2M)serum levels in patients with Alzheimer disease(AD).Methods Forty AD patients were taken into mini-mental state examination(MMSE)tests.Their IL-1β,IL-6 andα2M levels of serum were detected using enzyme-linked immunosorbent assay(ELISA)and then the results were compared with that of Parkinson’s disease(PD)and normal controls.The relationship between serum IL-1β,IL-6,α2M levels and MMSE score were analyzed in AD patients.In addition,the correlation between IL-1β,IL-6 andα2M were analyzed in AD group.Results①The serum IL-1βlevels significantly increased in the PD patients(P<0.05).There was a slight increase in the serum IL-1βlevels of the AD patients compared with healthy controls,the difference was not statistically significant(P>0.05).②The serum IL-6 levels significantly increased in the AD and PD patients(P<0.05),the IL-6 levels had no significant difference among the two groups(P>0.05).③Our data indicated no significant difference in AD and PD groups in the serumα2M levels(P>0.05).④The serum IL-6 levels were inversely correlated with MMSE scores(r=-0.281,P=0.016)in AD group.No correlation between serumα2M levels and other Inflammatory factors of AD(P>0.05).Conclusion These findings suggest that AD and PD are accompanied by a peripheral inflammatory response and that IL-6 may be associated with the degree of cognitive impairment of AD,IL-1βis not associated with the degree of cognitive impairment of AD,α2M is not be as a candidate marker for clinical diagnosis of AD.
