摘要
目的:研究酱香型白酒中生物活性物质(BSML)抗幽门螺杆菌代谢产物(Helicobacter pylori metabolite,HPM)诱发炎性损伤的作用及机制。方法:采用氯仿萃取及真空冻干获得BSML,GC-MS分析其主要成分,HPM诱导人脐静脉内皮细胞(HUVEC-12)建立炎性损伤模型;MTT法检测BSML和HPM对HUVEC-12细胞活性的影响;ELISA测定BSML处理细胞对相关炎症因子白细胞介素-4(Interleukin-4,IL-4)、白细胞介素-6(Interleukin-6,IL-6)、白细胞介素-8(Interleukin-8,IL-8)、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)分泌的影响,Western blotting检测BSML刺激细胞对NF-κB,MAPK信号通路的影响。结果:BSML可增强HUVEC-12细胞抗HPM导致的细胞活性降低,可显著降低HPM导致炎性因子IL-6、IL-8、TNF-α的分泌并促进抑炎因子IL-4的表达。Western blotting结果显示BSML可通过调控NF-κB,MAPK等信号通路发挥抗HPM诱发的炎性损伤。结论:酱香型白酒中含有的活性物质具有一定的抗炎活性,可通过抑制炎症因子IL-6、IL-8和TNF-α,促进抑炎因子IL-4的分泌及调控NF-κB、MAPK等信号通路发挥抗炎作用。
Objective:To study the effect and mechanism of BSML in Maotai-flavor liquor on anti-inflammatory injury induced by HPM.Methods:BSML was extracted by chloroform and freeze-dried in vacuum.The main components of BSML were analyzed by GC-MS.human umbilical vein endothelial cells(HUVEC-12)were induced by HPM to establish an inflammatory injury model.MTT method was used to detect the effects of BSML and HPM on HUVEC-12 cell activity.ELISA was used to detect the effects of BSML treated cells on the related inflammatory factors,IL-4 and IL-6,The effects of IL-6,IL-8,TNF-αsecretion and the effects of BSML stimulation on NF-κB,MAPK signaling pathway were detected by Western blotting.Results:BSML could enhance the anti HPM activity of HUVEC-12 cells,decrease the secretion of IL-6,IL-8,TNF-αand promote the expression of IL-4.Western blotting showed that BSML could inhibit HPM induced inflammatory injury by regulating NF-κB,MAPK and other signaling pathways.Conclusion:the active substances contained in Maotai-flavor liquor have certain anti-inflammatory activity,which can play an anti-inflammatory role by inhibiting the secretion of IL-6,IL-8 and TNF-α,promoting the secretion of IL-4 and regulating NF-κB,MAPK and other signal pathways.
