摘要
目的:应用瘤细胞悬液移植法建立SHZ-88大鼠乳腺癌细胞皮下移植瘤模型,并使用超声监测肿瘤生长特性。方法:选取雌性SD乳鼠20只,采用处于对数生长期的SHZ-88细胞悬液进行腋窝皮下接种,超声观察成瘤率、肿块大小、血供情况及声像图特征。结果:大鼠接种成功率为100%,生长良好;超声检查肿块为类圆形,内部不均匀低回声,边界较清,后方回声衰减,彩色多普勒扫查肿块内部及周边可见较丰富血流信号;3 w后切除肿瘤结节,大体观肿块成苍白色,多呈椭圆形,边缘呈分叶状改变,肿瘤血管较丰富,镜下肿瘤细胞大小形态不一,核大深染,异型性明显。结论:通过SHZ-88大鼠乳腺癌细胞接种于雌性SD乳鼠腋窝皮下,成功建立了大鼠乳腺癌移植瘤模型。该模型与人乳腺癌的组织学形态及生物学行为相似,且移植瘤的生长状况、回声类型及血供变化大体符合乳腺癌的一般超声表现特点,是较理想的影像学研究动物模型。
Objective:To establish subcutaneous transplanted breast cancer model in rat using SHZ-88 cell suspension,and monitor tumor growth characteristics by ultrasound.Methods:A total of 20 female SD rats were inoculated subcutaneously with SHZ-88 cell suspension in the logarithmic growth phase.Ultrasound was performed to measure the tumor formation rate,mass size,blood supply and sonographic features.Results:The success rate of inoculation was 100%,and the transplants growed well.Ultrasound examination indicated that the tumor was round with internal unevenness low echo.The boundary was clear with posterior echo attenuation.The color doppler showed there were abundant blood flow signals inside and around the mass.On 3 weeks after inoculation,the tumor was pale and mostly elliptical,with lobulated edges and abundant blood vessels.In addition,the cells were different in sizes and shapes,which had large and deep nuclei with obvious atypia under microscope.Conclusion:The transplanted breast cancer model using SHZ-88 in female SD rats has been established successfully.This transplanted tumor has similar characteristics of histological morphology and biological behaviors to human breast cancer.Moreover,the ultrasound characteristics of this transplant,including growth status,echo type and blood supply are generally consistent with breast cancer,which is an optimal animal model for imaging studies.
