五味子甲素对结肠炎大鼠氧化应激水平的调控作用 被引量:12

摘要 目的探讨五味子甲素对溃疡性结肠炎(ulcerative colitis,UC)大鼠结肠组织氧化应激水平的调控作用。方法用三硝基苯磺酸(Trinitrobenzenesulfonic acid,TNBS)/乙醇混合液灌肠复制结肠炎SD大鼠模型,大鼠随机均分为正常对照组、模型组、阳性对照组及五味子甲素低剂量(20 mg·kg^-1)、中剂量(40 mg·kg^-1)、高剂量组(80 mg·kg^-1),每组10只。灌胃给药,正常组与模型组同期分别给予等体积蒸馏水,阳性对照组给予150 mg·kg^-1美沙拉嗪(5-ASA),连续治疗7 d后取大鼠结肠并评测其结肠重量、结肠长度、肠重指数、病理切片以及选择最佳剂量组的结肠组织检测SOD,GSH-PX,NO等氧化应激指标。结果经治疗后,与模型组比较,五味子甲素中剂量组治疗效果最佳,肠重减轻、长度增加、肠重指数明显下降,炎症因子明显减少,GSH-PX,SOD,eNOS,iNOS和T-AOC的水平明显升高(P<0.01 or P<0.05),而NO及MPO水平显著降低(P<0.01 or P<0.05)。结论五味子甲素可能通过降低结肠组织中性粒细胞数量和NO含量以及升高SOD和GSH-PX的表达量缓解机体的氧化应激水平治疗实验性大鼠结肠炎。 Objective To explore the regulation of Schisandrin A on oxidative stress associated with colonic tissues in ulcerative colitis rats(UC).Methods The rat model of colitis was reconstituted with trinitrobenzenesulfonic acid(Trinitrobenzenesulfonic acid,TNBS)/ethanol mixture and rats were randomly divided into normal control group,model group,positive control group and low,middle or high dose(20,40,80 g·kg^-1)of Schisandra A groups,10 in each group.The normal group and the model group were given equal volume of distilled water for intragastric administration,the positive control group was given mesalazine(5-ASA)with the dose of 150 mg·kg^-1.After 7 days treatment,the colon was taken out and weighted.The colon length,intestinal weight index and pathological section were measured and we selected the optimal dose group’s colon tissue to detect SOD,GSH-PX,NO and other oxidative stress indicators.Results After treatment,compared with the model group,the Schisandra A medium dose group had the best therapeutic effect,the intestinal weight was reduced,the length was increased,the colon weight index was significantly decreased,and the inflammatory factors were significantly reduced.The levels of GSH-PX,SOD,eNOS,iNOS and T-AOC were significantly increased(P<0.01 or P<0.05),while the levels of NO and MPO were significantly lower(P<0.01 or P<0.05).Conclusion Schisandrin A treated experimental rat colitis maybe related to reducing the amount of neutrophils and the content of NO in colon tissue,increasing the expression levels of SOD and GSH-PX to alleviate oxidative stress levels in the body.
