摘要
对采自辽宁省沈阳市大豆生产田的大豆花叶病毒沈阳分离物(SMV-SY)进行了基因全序列的克隆及序列分析。经提取叶片总RNA,进行了RT-PCR鉴定。结果表明,其SMV-SY整条基因组由9 587个碱基组成,编码多聚蛋白的开放阅读框位于第132~9 335 nt,得到全基因序列上传至NCBI数据库,获得登录号为MK350280;BLAST比对结果显示,SMV-SY与其他分离物全基因组核苷酸序列之间的一致性在90.94%~96.86%;与国内外SMV不同分离物构建全基因组核苷酸序列系统进化树。分析表明,其与中国哈尔滨、江苏、以及伊朗和美国的分离物亲缘关系较近,与中国河北、重庆、南昌、广西、以及韩国和加拿大的分离物关系较远。
The complete genome of the Soybean mosaic virus Shenyang isolate(SMV-SY)that isolated from soybean field in Shenyang, Liaoning Province was cloned and sequenced. The SMV-SY contained 9587 nt, and the open reading frame encoding the polyprotein was located at 132 ~9 335 nt. The obtained complete genome sequence was submitted to the NCBI with the accession number MK350280. The result of the nucleotide BLAST analysis indicated that the identity between the complete genome sequence of SMV-SY and other isolates ranged from 90.94% to 96.86%. The phylogenetic tree was constructed with the complete genome sequence of domestic and foreign SMV isolates, showing that SMV-SY closely related to the isolates from Harbin and Jiangsu of China, Iran, the United States, and had a distant relationship with the isolates from Hebei, Chongqing, Nanchang, Guangxi of China, South Korea, and Canada.
