响应面分析法优化黄精多糖提取工艺及含量测定 被引量:11

Optimization of Polysaccharide Extraction from Polygonati Rhizoma by Response Surface Methodology and Its Content Determination

摘要 目的研究黄精多糖的水提醇沉工艺及含量测定方法。方法采用单因素试验及Box-Behnken Design方法,以提取物中的多糖含量为考察指标,优化最佳提取工艺;利用蒽酮-硫酸法对提取的多糖含量进行测定。试验设计的三因素中,液料比对黄精多糖含量影响极为显著,提取时间对结果有显著的影响,提取温度对结果影响不显著。结果黄精多糖的最佳提取工艺条件为:液料比20∶1(m L/g)、提取时间2 h,提取温度85℃,按照最佳提取工艺条件提取黄精中的总多糖,含量可达12. 50%。结论响应面法优化黄精多糖的提取工艺可靠,具有实用价值,可用于指导生产实践。 Objective Response surface methodology was used to investigate the optimization of water extraction and ethanol precipitation polysaccharide from Polygonati Rhizoma and determine its content in the optimum process.Methods Using Polygonati Rhizoma as test material,taking the content of polysaccharide as the response value,the optimum extraction process was optimized by single factor and response surface methodology.The polysaccharide content was determined by anthrone-sulfuric acid method.Results The results showed that the optimum processes for water extracting and ethanol precipitating of polysaccharide were as follows:the ratio of liquid to material 20:1(mL/g),extraction time 2 h,extraction temperature 85℃.The analysis of the response surface test showed that the ratio of liquid to material and extraction time was significant on the polysaccharide content,while extraction temperature had no significant effect on the polysaccharide content.Under the optimal condition,the content of polysaccharide from Polygonati Rhizoma was 12.50%.Conclusion This optimal extraction process could significantly improve the polysaccharides extraction rate.The experimental results were accurate and reliable.This study could provide reference data for developing products.
