摘要
目的探讨黄芪提取物对老年痴呆大鼠学习、记忆及海马组织中基质金属蛋白酶-2,9含量的影响。方法 2019年1月,通过向大鼠的侧脑室注射链脲佐菌素制备老年痴呆大鼠的动物模型,实验分成4组:对照组、模型组、黄芪提取物组及脑复康组。脑复康组和黄芪提取物组分别口服黄芪提取物和脑复康,对照组与模型组口服同样量的生理盐水,治疗时间为28 d。Morris水迷宫测定每组鼠的逃避潜伏期和穿台次数,免疫印迹实验测定各组大鼠海马组织中基质金属蛋白酶-2,9的含量。结果与对照组相比,模型组大鼠逃避潜伏期延长,而穿台次数减少(P<0.01),海马组织中基质金属蛋白酶-2,9相对含量明显上升(0.731±0.081、0.698±0.072,P<0.01)。与模型组相比,口服黄芪提取物的大鼠逃避潜伏期缩短,而穿台次数增加(P<0.01),海马组织中基质金属蛋白酶-2,9相对含量均显著下降(0.326±0.039、0.304±0.045,P<0.01)。结论黄芪提取物可降低老年痴呆大鼠海马组织中基质金属蛋白酶-2,9含量,有利于提高其学习与记忆功能。
Objective To explore the effects of Astragalus extract on learning and memory and the content of matrix metalloproteinase-2,9 in hippocampus of Alzheimers disease rats. Methods The animal model of Alzheimer’s disease was established by injecting streptozotocin into the lateral ventricle of rats on January 2019. The experiment animals were separated into four groups: control group,model group,Astragalus extract group and Naofukang group. Astragalus extract group and Naofukang group were given Astragalus extract and Naofukang respectively. The control group and the model group only took the same amount of sterile saline orally. The treatment durations were 28 days. Morris water maze was used to measure the escape latency and space exploration abilities of each group of rats. The content of matrix metalloproteinase-2,9 in hippocampus of rats in each group was determined by immunoblotting. Results Compared with the control group,the escape latency of model group prolonged while the number of space exploration was reduced(P<0.01),and the relative contents of matrix metalloproteinase-2,9 in hippocampus increased significantly(0.731±0.081,0.698±0.072)(P<0.01). Compared with the model group,the escape latency of Astragalus extract group shortened while the number of space exploration was increased(P <0.01),and the relative contents of matrix metalloproteinase-2 and 9 in hippocampus were significantly decreased(0.326±0.039,0.304±0.045)(P<0.01).Conclusion Astragalus extract can improve the learning and memory ability by reducing the content of matrix metalloproteinase-2,9 in hippocampus of rats with Alzheimers disease.
