摘要
研究构建大豆成花素GmFT5a基因启动子与pGreenII 0800双分子萤光素酶融合植物表达载体,农杆菌注射烟草,通过烟草体内瞬时表达活性试验分析长、短日照条件对GmFT5a启动子表达活性的影响,探讨GmFT5a受日长的表达影响。结果表明,经短日照处理的GmFT5a启动子表达活性高于长日照处理。说明短日照诱导GmFT5a启动子表达,进一步说明该基因为光周期调控因子。利用PLANTCARE在线预测工具分析GmFT5a基因启动子序列可知,GmFT5a启动子序列包含BOX 4、G-box、GT1-motif、Gap-box、chs-CMA1a光响应调控元件。由此说明GmFT5a基因可能通过启动子上光响应元件调控参与调控光周期。
In this study, a plant expression vector for the fusion of the promoter of the soybean florigen GmFT5a gene and pGreenⅡ 0800 bimolecular luciferase was constructed. Tobacco was injected with Agrobacterium, and the effects of long and short day conditions on the expression activity of GmFT5a promoter were analyzed by transient expression activity experiments in tobacco. The results showed that the expression activity of the GmFT5a promoter treated with short day light was higher than that of the GmFT5a promoter treated with long day light. It indicated that short-day sunlight could induce the expression of the GmFT5a promoter, further indicated that the gene was a photoperiod regulatory factor. The PLANTCARE online prediction tool was used to analyze the promoter sequence of the GmFT5a gene. The results showed that the GmFT5a promoter sequence contained elements of BOX 4, G-box, GT1-motif, Gap-box, and chsCMA1a light regulation. It indicated that the GmFT5a might participate in the regulation of the photoperiod through regulating the light-responsive elements on the promoter.
