虾青素对ApoE基因敲除小鼠动脉粥样硬化的降脂、抗氧化和抗炎作用研究 被引量:15

摘要 目的:研究虾青素对动脉粥样硬化(AS)的影响。方法:以高脂餐食喂养ApoE基因敲除小鼠造出AS模型;定期称体质量,在50、100、150 d时取血样,分析血清血脂指标[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)]和血糖水平;在150 d检查抗氧化指标:总抗氧化能力(T-AOC)、总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA);采用酶联免疫吸附实验(ELIAS)检查炎症因子IL-6、单核细胞趋化蛋白1(MCP-1)和C反应蛋白(CRP),采用苏丹红Ⅳ染色法检查动物主动脉斑块,采用Chrono-log血小板功能凝集计检测血小板聚集率。结果:虾青素治疗组TC、TG、LDL-C、血糖水平、MDA、IL-6、MCP-1和CRP明显降低(P<0.05),而HDL-C、T-AOC、T-SOD和GSH-Px明显高于高脂模型组(P<0.05);与高脂模型组比较,虾青素组斑块明显减小(P<0.05)。结论:虾青素对ApoE基因敲除小鼠动脉粥样硬化的发生和发展具有一定的保护作用,这可能与其降脂、抗氧化和抗炎作用有关。 Objective:To investigate the effect and possible mechanism of astaxanthin on the atherosclerosis(AS).Methods:ApoE-/-mice AS model were built through feeding high-fat diet.Blood was analyzed at 50 d,100 d and 150 d to monitor the lipid proile[total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)]and blood glucose.Antioxidant index containing total antioxidant capacity(T-AOC),total superoxide disumutase(T-SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)were determined at 150 d,inflammatory factor containing IL-6,monocyte chemoattractant protein-1(MCP-1)and C-reactive protein(CRP)in serum were investigated by enzyme-linked immunosorbent assay(ELISA).The aortic arch and brachiocephalic artery were analysed by SudanⅣstaining for plaque size and by morphometry for luminal occlusion at 150 d.The platelet aggregation was analyzed by Chrono-log platelet function aggregometer.Results:High fat group was successfully established atherosclerotic model.The serum TC,TG,LDL-C,blood glucose,MDA,IL-6,MCP-1 and CRP of astaxanthin group obviously was less than those of model group(P<0.05),while the level of HDL-C,T-AOC,T-SOD and GSH-Px was higher than those of model group(P<0.05).The aortic plaque size and platelet aggregation of astaxanthin group were both smaller than those of model group(P<0.05).Conclusion:The astaxanthin can effectively delay the development of AS of ApoE-/-mice,which may be contributed to the function of reducing lipid,oxidation resistance and anti-inflammatory of astaxanthin.
