摘要
高迁移率族蛋白B1(HMGB1)是一种高度保守的染色体结合蛋白。现在越来越多的研究表明它作为一种重要的损伤相关模式分子(DAMPs),经乙酰化、甲基化、磷酸化和氧化还原等翻译后修饰(PTMs)后与细胞DNA和蛋白质相互作用,从而成为炎症、免疫及肿瘤等疾病的生物标志物或参与疾病发展过程。因此,对于PTMs与HMGB1多种生物学活性之间复杂关系的研究逐渐受到重视,这也使HMGB1的PTMs作为疾病治疗的新靶点成为可能。本文就PTMs对HMGB1的结构、生物学效应的影响及其在疾病发展过程中的作用进行综述。
HMGB1 is a highly conserved chromosome-binding protein.Now more and more studies have shown that,as an important damage associated molecular patterns(DAMPs),HMGB1 interacts with cell DNA and protein after post-translational modifications(PTMs)by acetylation,methylation,phosphorylation and redox,thus then becoming a biomarker or involved in the development of diseases such as inflammation,immunopathy and cancer.Therefore,more and more attention has been paid to the complex relationship between PTMs and various biological activities of HMGB1,which makes PTMs of HMGB1 possible as a new target for disease treatment.This review summarizes the role of PTMs in the structure,biological effect of HMGB1 and in the development of diseases.
