黄葵素对1型糖尿病大鼠血清肌酐、尿素及尿微量白蛋白的影响 被引量:2

Effect of Abelmoschus on Serum Creatinine,Urea Nitrogen and Urine Microalbumin Indexes in Type 1 Diabetic Rats

摘要 目的探讨黄葵素对1型糖尿病(type1 diabetes mellitus,T1DM)大鼠肾功、尿微量白蛋白(urine microalbumin,UMALB)及肾组织病理变化的影响。方法选取SD大鼠为实验对象,随机分为空白组、模型组和实验组,应用链脲佐菌素对模型组和实验组进行造模。成模后每天给予实验组大鼠0.7g/kg体质量的黄葵素灌胃治疗,给予空白组和模型组同等剂量的蒸馏水,每周留取各组大鼠尿液测量UMALB含量,给药4周后处死,留取血液和肾脏标本。血液离心后取血清检测各组大鼠血清肌酐(creatinine,Cr)、尿素(urea nitrogen,UN)、胱抑素C(cystatinC,CysC)值,肾脏组织石蜡包埋切片进行苏木精-伊红(hematoxylin eosin,HE)染色,观察各组大鼠肾组织病理变化情况。结果黄葵素干预治疗4周后,实验组大鼠血清Cr、UN、CysC值分别为(36.19±4.93)μmol/L、(15.66±3.72)mmol/L、(0.13±0.04)mg/L,显著低于模型组各项指标(P<0.01);实验组UMALB的4周平均值为(7.48±7.37)mg/L,显著低于模型组(23.61±9.36)mg/L(P<0.01);HE染色结果可见实验组大鼠肾脏组织明显较模型组病理改变轻微,肾小球损伤较小。结论黄葵素可以改善T1DM大鼠肾脏功能,延缓糖尿病肾脏病(diabetic kidney disease,DKD)的发生发展,并具有排除其他治疗外的独立疗效。 Objective To investigate the effects of abelmoschus on renal function,urinary microalbumin and renal histopathological changes in rats with type 1 diabetes mellitus(T1DM).Methods SD rats were randomly divided into blank group,model group and experimental group.The model and the experimental group were established by streptozotocin.After the establishment of the model,rats in the experimental group were given 0.7g/kg body weight of abelmoschus by gavage every day,rats in the blank group and model group were given distilled water of the same dose,the urine of each group rats was collected weekly to measure the content of urinary microalbumin(UMALB),after 4 weeks of administration,the blood and kidney samples were executed,after blood centrifugation,the serum was taken to detect creatinine(CR),urea nitrogen(UN),Cystatin C(CysC)in each group,the kidney tissues were paraffin embedded and sectioned,HE staining was performed to observe the pathological changes of kidney tissues of rats in each group.Results After 4 weeks of treatment with abelmoschus,the serum Cr,UN and CysC values of experimental group rats were(36.19±4.93)mmol/L,(15.66±3.72)mmol/L and(0.13±0.04)mg/L,respectively,which were significantly lower than those of model group(P<0.01).The 4-week mean value of UMALB in the experimental group was(7.48±7.37)mg/L,which was significantly lower than that of model group(23.61+9.36)mg/L(P<0.01).HE staining showed that the pathological changes in the experimental group were slighter than those in the model group,and the glomerular damage was less.Conclusion Conclusion Abelmoschus can improve the kidney function of T1DM rats,delay the occurrence and development of diabetic kidney disease(DKD),and has an independent effect excluding other treatments.
