摘要
目的研究miR-192在肝细胞癌中的表达及其对迁移、侵袭的影响。方法选择2018年1月至2019年8月本院采集的110例原发性肝癌样本组织作为研究对象,经血清miR-192检测,分析其在肝癌中的表达情况,并研究miR-192与肝癌细胞迁移、增殖相关性。结果肝癌TNM分级为Ⅲ~Ⅳ级、肿瘤数量>1个、肿瘤>5 cm及有血管侵犯的肝癌血清miR-192高表达率,均低于TNM分级为Ⅰ~Ⅱ级、肿瘤数量为1个、肿瘤≤5 cm及无血管侵犯,差异均有统计学意义(均P<0.05);转染48 h后,肝细胞癌细胞增殖计数为(0.98±0.01),明显低于未转染组(1.48±0.02),转染72 h后,肝细胞癌细胞增殖计数为(1.21±0.02),明显低于未转染组(1.99±0.01),差异均有统计学意义(均P<0.05);miR-192表达水平与肿瘤分化程度、TNM分期、化疗敏感性有明显相关性,差异有统计学意义(P<0.05)。结论慢性病毒介导miR-192与肝癌细胞增殖、迁移能力密切相关,可抑制肝癌细胞生长增殖,肝癌程度越高则miR-192表达越低,可为肝细胞癌靶向诊治提供依据。
Objective To study the expression of miR-192 in hepatocellular carcinoma and its effect on migration and invasion.Methods A total of 110 primary liver cancer samples collected from our hospital from January,2018 to August,2019 were selected as study objects.The expression of miR-192 was analyzed by serum miR-192.The correlation of miR-192 with liver cancer cell migration and proliferation was studied.Results The high expression rates of serum miR-192 in the patients with TNM gradeⅢ-Ⅳliver cancer,>1 tumors,>5 cm tumor,and vascular invasion were lower than those in the patients with TNMⅠ-Ⅱtumor,1 tumor,≤5 cm tumor,and no vascular invasion(all P<0.05).48 hours after the transfection,the hepatocellular carcinoma cell proliferation count was(0.98±0.01),which was significantly lower than that,(1.48±0.02),in the non-transfection group(P<0.05).72 hours after the transfection,the hepatocellular carcinoma cell proliferation count was(1.21±0.02),which was significantly lower than that,(1.99±0.01),in the non-transfected group(P<0.05).The expression level of miR-192 was significantly correlated with tumor differentiation,TNM stage,and chemotherapy sensitivity(all P<0.05).Conclusion Chronic virus-mediated miR-192 is closely related to the proliferation and migration ability of liver cancer cells,and can inhibit the growth and proliferation of liver cancer cells.The higher the degree of liver cancer,the lower the expression of miR-192,providing a basis for targeted diagnosis and treatment of hepatocellular carcinoma.
