含Glypican3单克隆抗体电子顺磁共振成像诊断剂的合成与性能

Preparation and Property of Electron Paramagnetic Resonance Imaging Diagnostic Agent Based on Glypican3 Monoclonal Antibody

摘要 以5-苯氧乙酸乙酯联吡咯甲烷与5-醛基-异氮杂茚氧化氮自由基(FTMIO)为原料,在催化剂的作用下进行Lindsedy法成环反应与脱酯化反应,从而制备水溶性5-(1',1',3',3'-四甲基-异氮杂茚-2'-氧化氮自由基)-10,15,20-三[(4"-羧甲氧基)苯基]-卟啉(TmioTCPP)。将TmioTCPP与肝癌靶向基团磷脂酰肌醇蛋白聚糖3单克隆抗体(Glypican3-mAb)偶联,制备含Glypican3单克隆抗体电子顺磁共振成像诊断剂。对所合成诊断剂进行质谱和红外光谱等结构表征,进一步测试诊断剂的荧光、电化学与电子顺磁共振等性能。实验结果表明:所合成的诊断剂具有与TmioTCPP相似的荧光特性,以及与FTMIO相似的氧化还原性能。 A water-soluble 5-(1',1',3',3'-tetramethylisoindolin-2'-yloxyl-5'-yl)-10,15,20-tris(4"-carboethoxymethyleneoxyphenyl)-porphyrin(TmioTCPP)was synthesized by the Lindsey cyclization reaction between 5-(4'-carboethoxymethy-leneoxyphenyl)dipyrromethane and 5-formyl-1,1,3,3-tetramethylisoindolin-2-yloxyl(FTMIO).Subsequently,TmioTCPP was covalently attached with Glypican-3 monoclonal antibody(GPC3-mAb)to produce a tumor-targeting diagnostic agent for electron paramagnetic resonance imaging(TmioTCPP-GPC3-mAb).This diagnostic agent was further characterized by Fourier transform infrared spectroscopy and mass spectrometry.The fluorescence,cyclic voltammetry and electron paramagnetic resonance properties of the diagnostic agent were also evaluated herein.The diagnostic agent possessed similar fluorescence as TmioTCPP,and similar electrochemical and redox properties as FTMIO.
