摘要
目的建立肝母细胞瘤患者人源移植(PDX)模型,获得与患者肿瘤相似的并可传代的活体替代模型。方法获取肝母细胞瘤患者的新鲜肿瘤活性组织,无菌条件下4h内低温转运,台盼蓝染色法选取活性组织并剪切约3×3×3 mm3的组织块。将原代肿瘤细胞植入BALB/c裸鼠的肩胛骨前区和后肢的背面,当肿瘤的体积达到约800~1500mm^3时,取出一代肿瘤组织,将其中部分冻存,其余组织再次植入BALB/c裸鼠皮下传代。结果所获得PDX模型肿瘤组织与患者肿瘤组织细胞形态及结构均相似。结论采用患者肝母细胞瘤组织BALB/c裸鼠肿瘤移植等方法,可建立患者来源的肝母细胞瘤PDX模型,为进一步肿瘤药物筛以及精准治疗选提供了良好的动物模型。
Objective To establish patient-derived xenograft models of hepatoblastoma and obtain a living substitute model similar to the patient’s tumor. Methods Fresh tumor tissues were obtained from patients with hepatoblastoma and transported at low temperature within 4 hours under sterile conditions. The active tissues were selected by trypan blue staining and cut into about 3 × 3 × 3 mm3 tissue blocks. The primary tumor cells were implanted into the anterior area of scapula and the back of hind limbs of BALB/C nude mice. The first generation tumor tissues were taken out when the tumor volume reached about 800-1500 mm3,some of them were frozen, and the rest were implanted subcutaneously in of BALB/C nude mice. Results The cell morphology and structure of tumor tissue in PDX model were similar to those in patients. Conclusion The PDX model of hepatoblastoma from patients can be established by BALB/C nude mice tumor transplantation, which provides a suitable animal model for the research of anti-tumor drug screening and precise treatment.
