摘要
为确诊一例输入性卵形疟复发病例并鉴定其亚型,采集已开始服用青蒿素哌喹片的输入性卵形疟复发病例患者静脉血,通过薄血膜涂片吉姆萨染色镜检查看其形态特征,采用巢式PCR方法扩增其18S rRNA特异性基因,鉴定其疟原虫种类,再通过基因测序进行同源性比对和遗传进化分析。薄血膜涂片吉姆萨染色镜检结果显示,该患者血液红细胞中疟原虫已开始萎缩破裂,形态不典型,巢式PCR检测为卵形疟原虫强阳性。基因序列分析显示其序列与NCBI GenBank卵形疟Curtisi亚型序列同源性达99%。
In order to confirm a recurrent case of imported ovale malaria and identify its subtypes,the venous blood was collected of the patient with recurrent imported ovale malaria who had been administrated with artemisinin piperaquine tablets,and the morphological characters of P.ovale were examined by thin blood smear Giemsa staining.The 18S rRNA specific gene of Plasmodium in blood samples was amplified by Nested PCR,and identified to Plasmodium ovale.The homology and genetic evolution of the Plasmodium were analyzed by gene sequences.The malaria parasite in the erythrocytes of the patient had appeared atypical atrophy,rupture and showed strongly positive in nested PCR test with 99%identity of Plasmodium ovale Curtisi deposited in NCBI GenBank.
