摘要
【目的】了解西双版纳热带雨林植物叶际细菌群落结构。【方法】应用高通量测序分析方法,对随机7个调查采样点采集的39个无细菌症状的植物样品测序。【结果】草本样本细菌与木本植物细菌相比,共有的OTUs数量相似,木本植物细菌间差异的OTUs数量相对小于草本植物间的。在门(phylum)水平上主要检测注释到Cyanobacteria、Proteobacteria、Bacteroidetes,及未归类细菌门。在纲(class)水平上主要检测注释到Alphaproteobacteria、Bacteroidia、与Bacilli、Clostridia及未归类细菌纲。在属种水平上,Methylobacterium、Acinetobacte分布最广,Pseudomonas、Sphingomonas次之。Geobacter、Exiguobacterium、Nocardioides、Hymenobacter、Escherichia-Shigella、Pseudarthrobacter、Comamonas、Vogesella、Massilia与Aeromonas等细菌寄主范围窄,但是分布普遍。【结论】研究检测注释到植物病原细菌、益生细菌、其他细菌及未归类细菌。弄清了西双版纳叶围细菌群落结构,为开发利用潜在生物防治、医疗有益细菌,了解潜在植物致病细菌,认识细菌对热带雨林生态稳定性的角色与机制奠定了基础。
【Objective】The purpose of this study is to find out the bacterial community in the plant phyllosphere in Xishuangbanna.【Method】High-throughput microbial diversity sequencing and analysis methods were used to sequence the 39 collected plant samples from 7 randomly points.【Result】The common bacteria OUTs number from the herbal plants was similar to ones from the woody plants.The bacteria OTUs number from woody plants was relatively less than that from herbaceous plants.Cyanobacteria,Proteobacteria,Bacteroidetes and unassigned phylum were mainly detected and annotated at the phylum level.Alphaproteobacteria,Bacteroidia and Bacilli,Clostridia and unassigned class were mainly detected and annotated at the class level.At genus and species level,Methylobacterium and Acinetobacte were found the most widely distributed,followed by Pseudomonas and Sphingomonas.Geobacter,Exiguobacterium,Nocardioides,Hymenobacter,Escherichia-Shigella,Pseudarthrobacter,Comamonas,Vogesella,Massilia and Aeromonas had few hosts but widely distributed.【Conclusion】In this study,plant pathogenic bacteriium,good bacterium,other bacterium and unassigned bacterium were detected and annotated.The study clarified the bacterial community structure in Xishuangbanna,and laid a foundation for the development and utilization of potential biological control,medical beneficial bacteria,the understanding of potential plant pathogenic bacteria,and the understanding of the role and mechanism of bacteria in the ecological stability of tropical rainforest.
