银缕梅叶片中单宁提取纯化工艺的研究 被引量:3

摘要 为了进一步提高银缕梅的应用价值,运用了分子生物学中的液氮冷冻技术,研究了银缕梅中单宁的提取及纯化工艺。结果表明,银缕梅叶片单宁最佳提取纯化工艺为:60%丙酮,提取时间15min,料液比1∶10,提取温度50℃,在此条件下单宁酸提取量为72.00mg·g^-1。纯化工艺为:5 mg·mL^-1上样,流速为1.5mL·min^-1过AB-8型大孔树脂柱,最后用60%乙醇洗脱。 In order to improve its application value further,using the cryopreservation technology of liquid nitrogen in molecular biology,the extraction and purification process optimization of Tannins from leaves of Parrotia subaequalis was studied.The experiments showed that the extracting technique was optimum when the concentration of acetone was 60 wt.%,the extraction time was 15 minute,the ratio of solid-liquid was 1∶10 and the extracting temperature was 50℃.Under this condition,the highest amount of extraction of leaves of Parrotia subaequalis tannins was 72.00 mg·g^-1.The purification process indicated it was optimum while the sampling concentration was 5 mg·mL^-1,the current speed was 1.5 mL·min^-1 through macroporous resin column AB-8,and finally was eluted with 60 wt.%ethanol solution.
