摘要
目的检测Akt、p-Akt和Slug蛋白在三阴性乳腺癌及癌旁组织中的表达,分析三者与三阴性乳腺癌临床病理指标间的关系。方法应用免疫组织化学方法检测Akt、p-Akt和Slug蛋白在60例三阴性乳腺癌及癌旁组织中的表达。结果Akt、p-Akt及Slug蛋白在三阴性乳腺癌中的阳性表达率分别为76.67%、41.67%和56.67%,癌旁组织中Akt的表达率为6.67%,而p-Akt和Slug的表达率均为0;三阴性乳腺癌中Akt、p-Akt与Slug的表达与年龄及肿瘤组织学分级有关,年龄≤50岁患者的表达水平高于年龄>50岁患者;组织学分级越高,Akt、p-Akt与Slug的表达率越高。Akt、Slug与肿瘤大小有关,肿瘤越大,Akt、Slug的表达率越高。p-Akt的表达与有无淋巴结转移有关,有淋巴结转移组p-Akt的阳性率高于无淋巴结转移组;Slug与Akt、p-Akt在三阴性乳腺癌中呈正相关。结论在三阴性乳腺癌中,Akt、p-Akt和Slug蛋白均表达,三者的表达可能与三阴性乳腺癌的不良预后有密切关系,Slug与Akt、p-Akt在三阴性乳腺癌中呈正相关。它们可能通过EMT促进侵袭,转移和化疗耐药性。
Objective To investigate the expression of Akt,phospho-Akt and Slug proteins in triple-negative breast cancer(TNBC)and explore its relationship with clinical pathological indicators of TNBC.Methods The EnVision immunohistochemistry system was used to detect the expression of Akt,phospho-Akt and Slug proteins in 60 TNBC specimens and 60 specimens from normal tissues adjacent to breast cancer.The relationship between the expression of Akt,phospho-Akt and Slug,and clinical pathological features was analyzed.Results The positive expression rates of Akt,phospho-Akt and Slug were higher in TNBC tissues(76.67%,41.67%and 56.67%,respectively)than in normal tissues adjacent to breast cancer(6.67%,0%and 0%,respectively).The expression of Akt,phospho-Akt and Slug in TNBC was related to age and tumor histological grade,that is,the expression levels of patients aged≤50 years were higher than those of patients aged>50,and higher expression levels in the patients with higher histological grade.The expression of Akt and Slug was related to tumor size,and the larger the tumor,the higher the expression rate.The expression of phospho-Akt was related to the presence or absence of lymph node metastasis,and the positive rate was higher in the group with lymph node metastasis than in the group without lymph node metastasis.Slug was positively correlated with Akt and phospho-Akt in TNBC.Conclusion Akt,phospho-Akt and Slug were overexpressed in TNBC,which may play an important role in the poor prognosis and progression of TNBC.The expression of Slug had a positive correlation with Akt and phospho-Akt in the TNBC samples.They may promote the invasion,metasis and chemotherapy resistance through EMT.
